养心康片对心梗后心力衰竭模型兔血管紧张素Ⅱ、醛固酮和心肌病理形态的影响 被引量:3

Effects of Yangxinkang Decoction on Angiotensin Ⅱ,Aldosterone and Myocardial Pathological Morphology in Rabbits with Heart Failure after Myocardial Infarction

摘要 目的观察养心康片对心梗后心衰模型兔心功能、血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)和心肌病理形态的影响。方法应用结扎冠脉的方法建立心梗后心力衰竭兔模型,随机分为模型组和养心康组,并设立空白对照组,每组5只。造模后第3日开始给药,养心康组给予养心康片0.51 g/kg,灌胃每日1次。空白对照组和模型组给予等体积的蒸馏水,共4周。观察养心康片对模型兔心功能、AngⅡ、ALD和心肌病理形态的影响。结果模型组的左室舒张末容积(LVEDV)最高,与养心康组比较,差异有统计学意义(P<0.05);模型组左室射血分数(LVEF)、左室短轴缩短率(LVFS)值最低,与养心康组比较,差异有统计学意义(P<0.01);模型组的AngⅡ、ALD水平最高,与养心康组比较其差异有统计学意义(均P<0.05或P<0.01)。病理结果显示养心康组心肌纤维较模型组排列整齐,部分心肌纤维断裂,细胞间隙较空白对照组增宽,间质有少量炎症细胞浸润,心肌细胞部分水肿,局灶性心肌细胞坏死,较模型组明显改善。结论养心康片可以提高心梗后心衰模型兔的心功能,降低血清的AngⅡ、ALD水平,改善心肌病理结构。 Objective: To observe the effect of Yangxinkang Decoction on cardiac function,angiotensin Ⅱ (Ang Ⅱ ), aldosteroue (ALD) and myocardial pathological morphology in rabbits with heart failure after myocardial infarc- tion. Method: The rabbit model of heart failure was established with ligation of coronary artery. The experimental animals were randomly divided into the model group and Yangxinkang Decoction group, and the the blank control group was established.There were 5 rabbits in each group. The third day after modeling, Yangxinkang Decoction group were given Yangxinkang Decoction 0.51 g/kg, garage once a day. The blank control group and the model group were given equal volume of distilled water for 4 weeks. The effects of Yangxinkang Decoction on cardiac function, Ang U ,ALD and myocardial pathological morphology were Observed in rabbits. Results: Compared with that of Yangxinkang Decoction group, LVEDV of the model group was the highest, and the difference was statistically sig- nificant(P〈 0.05 ) ;the LVEF and LVFS values of the model group were the lowest, and the difference was statistical- ly significant (P〈 0.01 ) ;the levels of Ang II and ALD were the highest in the model group,and the difference was statistically significant (P〈 0.01 ). Pathological results showed that compared with the model group,the myocardial fibers of Yangxinkang Decoction group were neatly arranged;some of the myocardial fibers were ruptured ;the cell gap was wider than that of the blank control group; there was a little infiltration of inflammatory cells ; there was par- tial edema of cardiomyocytes,as well as necrosis of focal myocardial cells. All above was significantly improved, compared with the model group. Conclusion: Yangxinkang Decoction can improve the cardiac function of rabbits with heart failure after myocardial infarction, decrease the levels of Ang Ⅱ and ALD in serum and improve the my- ocardial pathological structure.
