摘要
目的观察哮喘模型大鼠肺组织白细胞介素-4(IL-4)及干扰素-γ(IFN-γ)的变化及防哮方的干预作用,探讨其防治哮喘的作用机理。方法将48只雄性SD大鼠按随机数字表法分为正常组,模型组,中药低、中、高剂量组及地塞米松组各8只。用卵蛋白(OVA)致敏激发建立慢性哮喘大鼠模型。收集支气管肺泡灌洗液(BALF)进行白细胞计数分类;逆转录—聚合酶链反应(RT-PCR)法检测肺组织中IL-4、IFN-γm RNA相对表达量。结果 1)与正常组比,模型组白细胞(WBC)总数及嗜酸粒细胞(EOS)百分率显著升高(P<0.05);与模型组比,中药低、中、高剂量组及地塞米松组WBC总数及EOS百分率明显降低(P<0.05);中药高剂量组与地塞米松组比,变化不明显(P>0.05)。2)与正常组比,模型组IL-4水平升高,IFN-γ水平降低(均P<0.05);与模型组比,中药低、中、高剂量组及地塞米松组,IL-4水平均降低,IFN-γ水平均升高(均P<0.05);中药高剂量组与地塞米松组比,IL-4水平较高(P<0.05),IFN-γ水平变化不明显(P>0.05)。结论防哮方可改善哮喘大鼠气道炎症,其作用机理可能与下调IL-4水平、上调IFN-γ水平、调节Th1/Th2免疫平衡有关。
Objective: To observe the effects of Anti-asthma Prescription on the changes of IL-4 and IFN-γ in asthmatic rats and explore the mechanism of prevention and treatment of asthma. Methods: 48 male SD rats were randomly divided into the normal group,the model group,low dose group,middle dose group,high dose group and dexamethasone group,with 8 rats in each group. The rat model of chronic asthma was induced by ovalbumin(O- VA). BALF was collected for the white blood cell differential count assay. RT-PCR was used to detect the ex- pression of IL-4 and IFN-γ mRNA in lung tissue. Results: 1 )Compared with the normal group,the total WBC and the percentage of EOS in the model group were significantly increased (P〈 0.05). The total WBC and the percentage of EOS in the low, middle and high dose groups were significant lower than those in the model group (P〈O.05). Compared with the dexamethasone group,the change of the high dose group was not obvious (P〉 0.05). 2)Compared with the normal group,the level of IL-4 was higher and the level of IFN-γ was decreased in the model group (P〈 0.05 ). Compared with the model group,the level of IL-4 were lower and the level of IFN-γ was increased in the low,middle and high dose groups (P 〈 0.05). Compared with the dexamethasone group,in high dose groups,the level of IL-4 were increased(P〈 0.05 ),and the level of IFN-γ had no significant difference (P 〉 0.05). Conclusion: Anti-asthmatic Prescription can improve airway inflammation in asthmatic rats. The mechanism may be related to down-regulation of IL-4 level,up-regulation of IFN-γ level and the regulation of Th 1/Th2 immune balance.
