摘要
目的观察黑骨藤追风活络胶囊对胶原诱导性关节炎(CIA)大鼠关节炎症的缓解作用及对血清白细胞介素-1β(IL-1β)、干扰素-γ(IFN-γ)、白细胞介素-15(IL-15)表达水平的影响。方法将40只雄性SD大鼠随机分为正常组、模型组、雷公藤多苷片组和黑骨藤追风活络胶囊组,每组10只,对除正常组外,其余3组建立CIA模型,于初次免疫后10 d开始,给各组大鼠进行相应药物灌胃处理以延缓炎症进展,并每隔7 d评定一次各组大鼠的关节炎指数(AI),给药21 d后处死大鼠检测各组大鼠血清中IL-1β、IFN-γ和IL-15的表达水平。结果正常组大鼠关节正常,而模型组关节红肿较为明显,AI指数评分较高(P<0.001),雷公藤多苷片组和黑骨藤追风活络胶囊组大鼠AI均较模型组有所缓解(P<0.01)。模型组大鼠血清IL-1β、IFN-γ、IL-15水平均较正常组升高(P<0.01),雷公藤多苷片组及黑骨藤追风活络胶囊组大鼠IL-1β、IFN-γ、IL-15水平均显著低于模型组(P<0.05或P<0.01)。结论黑骨藤追风活络胶囊可在一定程度上抑制CIA大鼠促炎因子分泌,缓解关节炎症,可作为临床上治疗类风湿关节炎的辅助用药。
Objective: To investigate the effect of Heiguteng Zhuifeng Huoluo Capsules on the alleviation of colla- gen-induced arthritis and the influence on the expression of serum IL-1β, IFN-γ and IL-15 in rat model. Methods: 40 male SD rats were divided into the normal group (group A),the model group (group B),Tripterygium wilfordii glycosides tablets group (group C) and HeigutengZhuifengHuoluo Capsules group (group D) at random, 10 rats in each. CIA model was established in group B ,C and D. The rats were given relative drugs 10 days after the first inoc- ulation to delay the progress of inflammation, and the arthritis index (AI) of rats in each group was evaluated every 7 days. After 21 days of drug administration, the rats were sacrificed and the levels of IL-1β, IL-15 and IFN-γ in the serum were measured to observe the effect of the drug on serum cytokines. Results: In the normal group, the joints of the rats were normal, but the joints in the model group were more red and swollen, and the AI index score was higher (P〈 0.001 ). Compared with the model group,the joints and AI in group C and D visibly alleviated (P〈 0.01 ). The levels of serum IL-1β, IFN-γ and IL-15 in the model group dramatically increased, compared with the normal group and there was a significant decline of the levels of these cytokines in group C and D, compared with group B. Con- clusion: Heiguteng Zhuifeng Huoluo Capsules can significantly inhibit the secretion of proinflammatory cytokines and reduce the degree of arthritis in CIA rats. It can be used as an auxiliary drug for the clinical treatment of rheuma- toid arthritis.
