摘要
【目的】确定细柄阿丁枫扦插生根适宜的生长调节剂配方,了解其扦插生根机制,从而提高扦插成活率,建立其扦插繁殖技术体系。【方法】采用3种植物生长调节剂(IAA、NAA和GGR)的4种质量浓度(150、300、500和800 mg/L)处理3年生细柄阿丁枫实生苗嫩枝进行扦插试验,以清水处理(CK)为对照,对插穗生根进程、生根性状(生根率、平均根数、平均根长)、生根进程中营养物质(可溶性蛋白质、淀粉、可溶性糖)含量和氧化酶活性(过氧化物酶(POD)、多酚氧化酶(PPO)、吲哚乙酸氧化酶(IAAO))进行动态测定。【结果】(1)植物生长调节剂对细柄阿丁枫扦插生根时间和生根性状具有极显著影响;(2)所有处理方法中,以300 mg/L NAA浸泡插穗2 h处理的扦插生根效果最佳,比CK提前12 d生根,生根率(78%)、平均根数(16.8条)、平均根长(4.27 cm)为CK的3.4倍、6.2倍和4.6倍;(3)插穗生根进程中营养物质含量和氧化酶活性各指标峰值或谷值出现的时间与不定根开始发生或大量发生的时间高度契合,这种节奏性的变化反映了生根进程与生理代谢之间的协同性。(4)300mg/L NAA处理枝条显著提高了氧化酶活性,加快了可溶性蛋白的积累速度和淀粉及可溶性糖的消耗速度。【结论】植物生长调节剂加快了营养物质的合成与代谢进程,提高了氧化酶的调节能力,所以加快了生根进程,并促进扦插苗生根。
【Objective】Determine the suitable formula and understand the mechanism of rooting Altingia gracilipes cuttings to improve cutting survival rate and establish a cutting propagation system.【Method】In this study,a total of 13 treatment groups using four different concentrations(150,300,500 and 800 mg/L) of IAA,NAA or GGR and one watertreated control(CK) were used to treat 3-year-old Altingia gracilipes softwood seedlings.The rooting process,rooting(rooting rate,average root number,average root length) and the dynamic changes in nutrient content(soluble protein,soluble starch,soluble sugar) and oxidase activity(peroxidase(POD),polyphenol oxidase(PPO),indoleacetic acid oxidase(IAAO)) were observed during the rooting process of Altingia gracilipes cuttings.【Result】(1)Plant growth regulators had a significant influence on rooting time and rooting of Altingia gracilipes cuttings.(2)Rooting of cuttings in the treatment with 300 mg/L NAA for 2 h were the earliest to exhibit the rooting process,12 d ahead of CK,and the rooting rate(78%),average root number(16.8),and average root length(4.27 cm),were,3.4,6.2 and 4.6 times that of CK.(3)Dynamic changes in nutritive material content and oxidase activity in the process of Altingia gracilipes rooting from the adventitious root induction period to the early adventitious root formation,than the adventitious root formation stage,indicated that the peak or valley value of each indicator appeared at the same time as the beginning time of the early adventitious root or adventitious root formation stage,which reflected the synergy between the rooting process and physiological metabolism.(4)The 300 mg/L NAA treatment significantly improved oxidase activities,the accumulation of soluble proteins.【Conclusion】Plant growth regulators sped up the process of synthesis and metabolism of nutrients and rooting,and improved the ability to regulate endogenous oxidase.
