星点设计-效应面法优化三氧化二砷纳米粒制备工艺 被引量:2

摘要 研究As_2O_3聚氰基丙烯酸正丁酯纳米粒(As_2O_3-PBCA-NP)的制备工艺。采用乳化聚合法制备纳米粒,以Poloxamer 188、α-BCA和As_2O_3用量为考察因素,以包封率、载药量和平均粒径及归一值(OD)值为考察指标,采用星点设计-效应面法,对结果进行多元线性回归及二项式拟合,预测较佳工艺条件。结果显示,星点设计-效应面法得到的优化处方制备的样品包封率为87.61%,载药量为1.39%,平均粒径为195.6nm,形态为圆形,分散均匀。星点设计-效应面法优化As_2O_3-PBCA-NP的制备工艺可行简便。 To investigate the preparation of arsenic trioxide polybutylcyanoacrylate nanoparticles (As203-PB- CA-NP).The nanoparticles were prepared through emulsion polymerization method. Poloxamcr 188,ct-BCA and As203 were considered as factors and entrapment efficiency, drug loading, average diameter and OD were chosen as evaluation parameters.In order to predict the optimal prescription, central composite design (CCD) and response surface methodology (RSM) were conducted to analyze the results by multi-linear regression and second-order polynomial equation. According to the optimal formulation, the entrapment efficiency, the drug loading and the av- erage diameter of the prepared nanoparticles were 87.61%, 1.39%, 195.6nm,respectively.The prepared As203-PB- CA-NP were uniformly distributed and in round shape.Combined of CCD and RSM was a convenient and feasible preparation method.
