Advances in molecular targeted therapy of pancreatic cancer

摘要 胰腺癌是恶性程度极高的一种消化系统恶性肿瘤,是中国恶性肿瘤的主要死亡原因之一。胰腺癌患者的预后很差,5年生存率低于5%。尽管吉西他滨作为胰腺癌患者的一线治疗方案,但是生存期仍无法超过6个月。目前,分子靶向治疗因其特异性及对肿瘤细胞高效的杀伤性而被认为是治疗胰腺癌潜在的有效方案。人们希望通过分子靶向治疗获得更佳的治疗效果。本文以临床试验数据为基础,主要从表皮生长因子受体抑制剂、胰岛素样生长因子受体抑制剂、血管内皮生长因子抑制剂、丝裂原活化蛋白激酶信号通路抑制剂以及间质表皮转化因子信号通路抑制剂这几个方面,阐述胰腺癌分子靶向治疗进展。 Objective: Pancreatic cancer is a highly aggressive gastrointestinal cancer and remains one of the leading causes of cancer-related mortality in China. Patients with pancreatic cancer have a poor prognosis with a 5-year survival of less than 5%. Despite using gemcitabine as first-line therapy for patients with pancreatic cancer, the survival does not exceed 6 months. At present, molecularly targeted therapeutics is regarded as potentially effective in the treatment of pancreatic cancer owing to its specificity and cytotoxicity. It is hoped that more favorable results can be obtained by using molecular targets. This review summarizes the targeted therapy of pancreatic cancer based on the relevant clinical trial data, including several aspects of epidermal growth factor receptor inhibitors, vascular endothelial growth factor inhibitors, mitogen- activated protein kinase signaling pathway inhibitors and cellular- mesenchymal to epithelial transition factor signaling pathway inhibitors.
