滇西亚种树鼩实验性酒精肝损伤研究 被引量:2

摘要 【目的】为了探讨酒精对滇西亚种树鼩肝脏的影响。【方法】试验将40只滇西亚种树鼩随机分为酒精组30只和对照组10只。试验树鼩灌服15 d时,灌服供试品后1 h、2 h、3 h、12 h时心脏采血EDTA-K2抗凝后测定血液中乙醇浓度。分别在灌服30 d、60 d和90 d时,禁食12 h心脏采血分离血清后测定肝功能生化指标,数据采用SPSS软件统计分析。树鼩灌服供试品后30 d、60 d、90 d时分别取肝脏使用分析天平称量、计算肝指数并作HE染色、Masson染色镜检和电镜观察。【结果】酒精组树鼩灌服酒精后1 h、2 h、3 h、12 h血液中乙醇质量浓度分别为153.94±11.25、165.84±7.10、153.81±5.15、0 mg/d L,对照组树鼩血液中乙醇浓度均为0 mg/d L。酒精组树鼩30 d和90 d时测定的ALT(181.65±39.86 U/L和124.15±31.24 U/L)显著高于对照组(79.73±10.71 U/L)(P<0.05),酒精组树鼩90 d测定的AST(234.92±13.76 U/L)显著高于对照组(200.10±20.08 U/L)(P<0.05)。酒精组树鼩30 d、60 d、90 d肝指数(0.045 2±0.015 4、0.045 5±0.019 8、0.042 3±0.007 4)与对照组(0.042 7±0.008 9)比较差异不显著(P>0.05)。HE染色镜检和电镜观察到酒精组树鼩60 d和90 d时肝细胞出现了病理变化。【结论】酒精对树鼩的肝脏能够产生毒性损伤作用,树鼩灌服酒精后血液中乙醇浓度出现高峰时间点与人较为接近。 [ Purpose ] In order to investigate the effect of alcohol on the liver of Tupaia belangeri chinensis and the ethanol concentration assay in blood. [ Method] Tested 40 tree shrews were ran- domly divided into ethanol group ( n = 30) and control group ( n = 10) in the experiment. Alter the administration for 15 days and EDTA-K2 anticoagulation, the concentration of ethanol in the blood was determined at 1 , 2, 3 and 12 h. At 30, 60 and 90 days after fasting 12 hours, the blood biochemical parameters of liver function were measured. The data were analyzed by SPSS software. The liver of the tested shrew was weighed using a Mettler Toledo analytical balance at 30, 60 and 90 days after the administration and then calculated the liver index. The liver was test- ed by HE staining, Masson staining and electron microscopy. [ Results] The ethanol concentra- tion in the blood of tree shrews was 153.94 ± 11.25, 165.84 ±7. 10, 153.81± 5. 15, 0 mg/ dL at 1, 2, 3 and 12 h after alcohol administration, respectively. The ethanol concentration in blood of the control group was 0 mg/ dL. ALT in the experiment group ( 181.65 ±39.86 u/ L and 124. 15 ± 31.24 U/L) were significantly higher than those in the control group (79.73± 10.71 U / L) at 30 days and 90 days (P 〈 0.05), and AST in the experiment group (234.92 ±13.76 U / L) was significantly higher than that in the control group (200. 10± 20.08 U / L) ( P 〈 0.05 ). The liver index was not significantly different ( P 〉 0.05 ) in the ex- periment group (0. 0452 ± 0. 0154, 0. 0455 ± 0. 0198, 0. 0423 ±0. 0074) and the control group (0. 0427 ±0. 0089) at 30, 60 and 90 days . HE staining and electron microscopy showed that hepatic cells showed pathological changes at 60 d and 90 d of alcohol group. [ Conclusion] Alcohol could damage the liver of T. belangeri ehinensis and the peak time of alcohol concentration in blood of T. belargeri chinensis was similar to that of human.
