摘要
目的:应用高效液相色谱法测定还脑益聪方中α-细辛醚、β-细辛醚和细辛醛的含量。方法:采用Agilent Extend-C18(4.6 mm×250 mm,5μm)色谱柱,用甲醇-水(含0.1%甲酸)梯度洗脱,检测波长265 nm,柱温30℃,进样量20μL,流速为1 m L/min,分别检测还脑益聪方中α-细辛醚、β-细辛醚和细辛醛含量,并用石菖蒲样品作对照。结果:石菖蒲样品和还脑益聪方中α-细辛醚的含量分别为1.274 2,3.489 1 mg/g,β-细辛醚含量分别为7.630 1,35.836 0 mg/g,细辛醛含量分别为0.833 7,7.808 3 mg/g。结论:还脑益聪方中α-细辛醚、β-细辛醚和细辛醛含量分别是石菖蒲样品的2.7倍、4.7倍和9.4倍,提示整方配伍后可提高三者的溶出率。
Objective:To determine the content of α-asarone,β-asarone and asarone in Huannao Yicong formula by HPLC. Methods:The chromatographic separations were performed on an Agilent Eclipse XDB-C18 column(250 mm×4.6 mm i.d.,5 μm) with 30 ℃. The mobile phase consisted of methanol and 0.1% formic acid water. The wavelength of detector was set at 265 nm,the flow rate was 1.0 m L/min. The content ofα-asarone,β-asarone and asarone was detected respectively. Results:The content of α-asarone in Rhizoma Acori Tatarinowii and HNYCF was 1.274 2 and 3.489 1 mg/g,the content of β-asarone was 7.630 1 and 35.836 mg/g,the content of asarone was 0.833 7 and 7.808 3 mg/g. Conclusion:The contents of α-asarone,β-asarone and asarone in HNYCF were 2.7,4.7 and 9.7 times than in Rhizoma Acori Tatarinowii,which indicated that the dissolution rate of the three could be increased after compatibility.
