水貂肠炎细小病毒及犬瘟热病毒灭活后的联合免疫效果 被引量:2

Combined immune efficiency of inactivated mink enteritis virus and canine distemper virus in rats

摘要 目的探讨水貂肠炎细小病毒(mink enteritis virus,MEV)和犬瘟热病毒(canine distemper virus,CDV)灭活后的联合免疫效果。方法用甲醛灭活MEV-L和CDV-L弱毒株,固定MEV-L的血凝效价为2~6,与CDV-L病毒液(CDV-L TCID_(50)10^(-5.29)/mL)分别按1∶1(Ⅰ组)、1∶2(Ⅱ组)、1∶3(Ⅲ组)的量混合,加入10%氢氧化铝胶,充分混匀后免疫大鼠,均为1 m L/只。分别于免疫前及免疫后第7、14、30、60、90天采血,分离血清,采用血凝抑制(hemagglutination inhibition,HI)试验和间接ELISA检测血清MEV抗体效价,血清中和试验和间接ELISA检测血清CDV抗体效价。结果大鼠在免疫后7 d时产生MEV和CDV抗体,30 d时抗体效价最高,随后下降,90 d时抗体效价仍较高;Ⅰ组CDV抗体效价较其他两组高。结论3组不同比例的CDV和MEV联合免疫大鼠均可产生较高的抗体水平,其中以1∶1比例的免疫效果最佳。 Objective To investigate the combined immune efficiency of inactivated mink enteritis virus(MEV)and canine distemper virus(CDV)in rats.Methods Attenuated MEV-L and CDV-L strains were inactivated with formaldehyde.The MEV-L at a hemagglutination(HA)titer of 2-6 were mixed with 10-5.29)TCID50/m L CDV-L at ratios of 1∶1(groupⅠ),1∶2(groupⅡ)and 1∶3(groupⅢ)respectively,added with 10%aluminum hydroxide adjuvant and immunized to rats,1 m L for each.Serum samples were collected before and on days 7,14,30,60 and 90 after immunization,and determined for MEV antibody titer by hemagglutination inhibition(HI)test and indirect ELISA,and for CDV antibody titer by serum neutralization test and indirect ELISA.Results MEV and CDV antibodies were induced7 d after immunization,of which the titers reached the peak values 30 d then decreased,while were remained at high levels 90 d after immunization.However,the CDV antibody titer in groupⅠwas higher than those in the other two groups.Conclusion Combined immunization with CDV and MEV at various ratios induced high antibody levels in rats.However,the immune efficiency of the mixture at a ratio of 1∶1 was satisfactory.
