针刺预处理对心梗后心衰大鼠心肌纤维化影响的实验研究 被引量:8

摘要 目的:探讨针刺预处理对心梗后心衰大鼠的心肌保护作用及其抗纤维化作用的相关机制。方法:45只雄性Wistar大鼠随机分为3组:假手术对照组、心梗后心衰模型组、针灸预处理组,建立大鼠心肌梗死后心衰模型,针刺预处理组于心梗后连续4周电针双侧肢体内关穴处理,4周后,超声心动图评价各组大鼠心功能;HE染色及天狼猩红染色测定左心室心腔大小及心肌纤维化情况;实时荧光定量PCR测定心肌组织微小RNA29a(miR29a)表达情况;蛋白免疫印迹法测定心肌纤维蛋白III型胶原蛋白(COL3A)表达情况。结果:心功能检测结果显示针刺预处理能够有效改善心梗后心衰大鼠左心室的舒张功能与收缩功能;HE染色及天狼猩红染色示:针刺预处理有效减小左心室心腔大小,且心室梗死区边缘纤维化面积比值较模型组小(P<0.01);qPCR示:相较于模型组,针刺预处理显著提升了miR-29a的表达量(P<0.05);Westernblot结果显示,针灸预处理显著降低心梗后心衰的胶原纤维蛋白COL3A的表达量(P<0.05)。结论:针刺预处理对心梗后心衰具有保护作用,其机制可能与抗心肌纤维化相关。 Objective: To research the protective effect of electroacupuncture preconditioning on myocardial injury in patients with heart failure after myocardial infarction and its mechanism of anti fibrosis. Methods: 45 male Wistar rats were randomly divided into 3 groups: sham group (control) , model group(UT) and electroacu- puncture preconditioning group(EP) , to establish a rat model of heart failure after myocardial infarction, the control group received sham operation, the UT group without any treatment after myocardial infarction , the EP group received 4 weeks of electroacupuncture treatment after myocardial infarction , echocardiography was used to evaluate the cardiac function of rats ; left ventricular cardiac cavity size was determined and myocardial fibro- sis was observed by HE staining and Sirius red staining;the expression of miR29a was detected by real -time flu- orescent quantitative PCR; Western blotting was used to detect the expression of myocardial fiber protein. Results: Electroacupuncture preconditioning could effectively improved the diastolic function and systolic function of left ventricular after myocardial infarction ; HE staining and Sirius red staining results showed that electroacupuncture preconditioning could effectively reduced the size of the left ventricle, ventricular infarction area and the edge of fibrosis area ratio compared with the UT group (P 〈 0.01 ) ; qPCR results showed : com- pared to UT group, electroacupuncture pretreatment significantly increased the expression of miR- 29a ( P 〈 0. 05 ) ; Western blot results showed that electroacupuncture pretreatment significantly decreased the expression of heart failure after myocardial infarction, its mechanism may be associated with inhibiting myocardial fibrosis.
