摘要 目的:观察黄枇烟吸入对肺癌小鼠肿瘤生长的抑制作用及其对去氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)表达及肿瘤微血管密度(MVD)的影响。方法:建立小鼠Lewis肺癌移植瘤模型,实验分为模型组、黄枇烟吸入低剂量组、高剂量组和顺铂组,每组10只。各组给药2周后,处死大鼠,取瘤称重,计算抑瘤率;免疫组化法检测瘤组织HIF-1α、VEGF的表达,光镜观察病记录MVD。结果:与模型组比较,黄枇烟吸入低剂量组、高剂量组和顺铂组均能够抑制肿瘤生长,而以化疗药顺铂组作用最强,黄枇烟吸入高、低剂量组次之。各给药组均能抑制肿瘤血管HIF-1α、VEGF因子的表达以及降低MVD计数,以顺铂组作用最明显,黄枇烟吸入高、低剂量组次之。结论:黄枇烟吸入可抑制肺癌小鼠肿瘤生长,其作用机制与下调HIF-1α、VEGF的表达及抑制肿瘤血管生成有关。 Objective: To observe the inhibition effect of Huangpi Smoke on lung cancer growth of mice and influences on HIF - 1 α, VEGF expressions and MVD in tumor tissues. Methods: Mice with Lewis cancer were built, experiment was divided into model group, Huangpi Smoke high and low dose groups, cisplatin group, each group with 10 mice. Mice of each group were given the corresponding medicine for 2 weeks, then taking out tumor tissues, weighing, calculating tumor inhibition rate, HIF - 1α, VEGF expressions in tumor tissues were determined by immunohistochemistry and MVD was observed by optical microscope. Results: Lung cancer growth of mice in Huangpi Smoke high , low dose groups and cisplatin group were inhibited, the tumor weight of the a- bove three groups were significantly lower than that of model, the tumor inhibition rate of cisplatin group was the highest. The HIF -1α, VEGF expressions in tumor tissues of Huangpi Smoke high , low dose groups and cispla- tin group were inhibited and MVD number was reduced, the effects of cisplatin group were the best, Huangpi Smoke group was the second. Conclusion : Huangpi Smoke can inhibit cancer growth, down - regulating HIF - 1α, VEGF expressions in tumor tissues and inhibiting MVD were part of the mechanism.
