摘要
CRISPR(Clustered regularly interspaced short palindromic repeats,规律成簇的间隔短回文重复)平台是出色的基因组编辑工具,其中新型核酸酶Cas13a(CRISPR-associated protein 13a)既可编辑DNA,也可编辑RNA,这一新平台可实现DNA或RNA的摩尔灵敏度以及单碱基错配的特异性检测,同时拓展用于病毒与细菌的精细检测(甚至是寨卡与登革热的近亲病毒株)、癌症早期监测和遗传性疾病等治疗;其需要设计少,技术操作简单,具有广泛的应用潜力,是基因组编辑的又一项卓越成果。综述了Cas13a系统的最新进展。
The CRISPR(clustered regularly interspaced short palindromic repeats) platform is an excellent genome editing platform, in which, the novel nuclease Cas13 a(CRISPR-associated protein 13 a) can edit DNA and RNA. This new platform could fulfill DNA or RNA detection with molar sensitivity and single-base mismatch specificity, and meanwhile be extended to fine detect of viruses(even Close relatives of Zika and Dengue strains) and bacteria, monitor early cancer, with the simple technical manipulation and a wide range of application potentials, which could be another excellent tool of genomic editing.Here the current progress about Cas13 a system was overviewed.
