摘要
目的:探讨早期生长反应因子-1(Egr-1)DNA酶(EDRz)对大鼠腹主动脉瘤(AAA)形成的抑制作用,进而证实EDRz治疗腹主动脉瘤的可行性。方法:对照组为弹力蛋白酶灌注的腹主动脉瘤组(AAA组),实验组为弹力蛋白酶灌注后经EDRz治疗的EDRz组。分别术前,术后28d测量腹主动脉直径。应用q-PCR方法检测动脉瘤中早期生长反应因子-1 mRNA的表达,结果进行统计学分析。结果:EDRz组动脉直径明显小于AAA组(AAA组:3.11±0.103,EDRz组:2.27±0.096,P<0.05)。与AAA组相比,EDRz组Egr-1mRNA的表达明显降低(AAA组:1.094±0.066,EDRz组:0.603±0.0448,P<0.05)。结论:Egr-1 DNA酶(EDRz)能特异性的降低Egr-1基因的表达并能够抑制腹主动脉瘤的形成与发展。
Objective:To investigate the inhibiting effect of( Egr-1) DNase( EDRz) on the formation of abdominal aortic aneurysm( AAA) in rats so as to confirm the feasibility of EDRz in the treatment of abdominal aortic aneurysm.Methods:Abdominal aortic aneurysm group( AAA group) taken as control group were injected by elastase.EDRz group taken as experimental group were treated with EDRz after perfusion of elastase.The diameter of the abdominal aorta was measured on the 28 th days before and after the surgery.The expression of early growth response factor-1 mRNA in aneurysms was detected by q-PCR,and the results were statistically analyzed.Results:The arterial diameters in EDRz group were significantly smaller than those in AAA group( AAA group:3.11± 0.103,EDRz group:2.27 ± 0.096,P < 0.05).Compared with AAA group,the expression of Egr-1 mRNA in EDRz group was significantly lower than that in AAA group( AAA group:1.094 ± 0.066,EDRz group:0.603 ±0.0448,P < 0.05).Conclusion:Egr-1 DNAase( EDRz) can specifically reduce the expression of Egr-1 gene and inhibit the formation and development of abdominal aortic aneurysm.
