摘要
LOG基因编码5′-核糖单磷酸水解酶,将细胞分裂素的前体物质直接转化成具有生理活性的细胞分裂素,在细胞分裂素合成途径中具有重要作用。为深入研究大白菜LOG基因家族的功能,利用生物信息学方法对大白菜基因组中LOG基因家族成员进行了鉴定,并对其基因组信息、蛋白质生理生化特征、基因结构、保守结构域、系统进化等方面进行了研究。结果表明:在大白菜基因组中共鉴定出17个LOG基因,可以聚类到7个分支A^G中;LOG基因在大白菜10条染色体上呈现不均匀分布,在第2和4号染色体上分布相对较多;对大白菜LOG蛋白氨基酸序列进行了多重比对,比对结果表明LOG蛋白的氨基酸中均具有一致序列为MHxRKAxMx(5)FxALPGGYGTxxEE的LOG蛋白特异的LDC motif;通过MEME软件对大白菜LOG蛋白motif的预测发现了8个比较保守的motif。研究结果为大白菜LOG基因功能的研究奠定了基础。
LOG genes encode riboside 5′-monophosphate enzymes,which could directly transform cytokinin precursors into cytokinins with physiological activities,thus acting essential roles in the biosynthetic pathways of cytokinins.To further research the LOG gene family functions of Chinese cabbage,bioinformatics tools were conducted to identify the LOGgene family members in the Chinese cabbage genome,and genome annotation,physiological and biochemical characteristics of proteins,gene structures,conserved domains and phylogenetic trees were also analyzed.The results showed that 17 LOGgenes were identified in the whole genome level,which could be clustered into 7 clades,A-G;LOG genes were unevenly distributed in the 10 chromosomes,among which chromosome 2 and chromosome4 contained relatively more LOG genes.Multiple alignment of LOG protein sequences were conducted and results indicated the consensus sequences of MHxRKAxMx(5)FxALPGGYGTxxEE,which was specific to LDC motif in LOG proteins.Besides,8 conserved motifs were predicted among LOG proteins by MEME analysis.These results established certain foundation for LOGgene functions study in Chinese cabbage.
