miR-940在调控胃癌细胞生长和迁移行为机制的研究 被引量:2

摘要 为探究micro RNA调控胃癌细胞生长和迁移等生物学行为的机制,本研究从基因表达综合数据库GEO下载7例胃癌micro RNA芯片数据,利用Qlucore Omics Explorer(QOE)3.2分析差异microRNA,筛选出miR-940做后续分析。本研究采用Real-time PCR方法检测miR-940在37例胃癌组织及胃癌细胞是否存在差异表达,并在胃癌细胞MKN-45中过表达miR-940,通过MTT、流式细胞术及划痕法检测胃癌细胞的细胞周期、细胞增殖及迁移能力。此外,我们利用双荧光素酶系统确定miR-940的靶基因,并用Western blotting法检测该靶基因在胃癌细胞中的表达水平。研究结果表明,miR-940在胃癌组织及胃癌细胞中的表达水平均显著上调,过表达miR-940可使胃癌细胞增殖能力加快,细胞周期缩短,增殖活性明显增高,迁移能力提升。ZNF191作为miR-940靶基因,在胃癌细胞中呈现低表达。根据以上研究结果可得出结论,miR-940在胃癌发生中高表达,可以促进胃癌细胞生长、提高胃癌细胞迁移能力,并且可能是通过调控ZNF191基因来发挥作用,这为理解胃癌的发生机制提供了理论依据。 In order to explore the regulation mechanism of micro RNA to growth and migration in gastric cancer cell, 7 gastric cancer micro RNA expression profile microarrays from GEO were downloaded, and QOE 3.2 was used to analyze differentially expressed micro RNAs and finally selected miR-940 for subsequent analysis. In this research, it was verified whether miR-940 differentially expressed in 37 gastric cancer tissues and cells through the real-time PCR, then miR-940 was overexpressed in gastric cancer cells. MTT, flow cytometry and scratch method were used to detect gastric cancer cell cycle, cell proliferation and migration ability. Dual-luciferase system and western blotting were also applied to detect miR-940 target gene and its expression level in gastric cancer cells.The results indicated that the expression level of miR-940 in gastric cancer tissues and cells increased significantly, the overexpression of miR-940 could increase gastric cancer cell proliferation, shorten cell cycle,increased proliferation activity significantly, as well as improved the migration ability. Moreover, ZNF191, the target gene of miR-940, also showed lower expression level in gastric cancer cells. Based on above results, it concluded that miR-940 was highly expressed in gastric cancer and could promote gastric cancer cell proliferation and migration ability, and exerted its effects through the regulation of ZNF191 possibly. It might provide theoretical basis for gastric cancer formation.
