摘要
以取自华东酒庄、烟台酒堡、宁夏葡萄园的25个葡萄(Vitis vinifera)品种为试材,对葡萄叶片进行DNA提取,选用4对核心引物进行PCR扩增,扩增产物进行毛细管电泳,所得微卫星等位基因数据与瑞士葡萄数据库进行比对,来鉴定25个葡萄品种。结果表明:利用4对基本核心引物对位于不同染色体上的SSR位点进行PCR扩增,对扩增产物进行拼接,通过得到的微卫星等位基因与葡萄数据库进行比对,可以准确鉴别19个酿酒葡萄品种,其他6个品种的3对引物得到的微卫星数据与数据库一致,其品种的鉴定需进一步实验确定。
By technology of SSR,25 cultivars which were taken from chateau of Eastern China,chateau of Yantai and vineyard of Ningxia were identified. Extracting DNA of the grape leaf,performing PCR amplification with 4 core primers and the expanded product was carried on capillary electrophoresis. The microsatellite allele data were compared in the Swiss grape database and identified 25 grape cultivars.The results showed that SSR locus on different chromosomes were amplified by four pairs of basic core primer and the amplification products were jointed. 19 wine grape cultivars could be identified by comparing microsatellite alleles with the grape database. The microsatellite allele data of the rest 6 cultivar was consistent with database and the identification of the 6 cultivars needed further experiment.
