蛤蚧不同提取液对衰老模型小鼠免疫功能的影响 被引量:7

摘要 目的探讨蛤蚧不同提取液对D-半乳糖诱导衰老小鼠免疫功能的影响。方法选择健康昆明种小鼠50只,随机分为正常对照组、模型对照组、维生素E组、乙醇提取液组和仿生酶解液组。正常对照组小鼠皮下注射生理盐水,其余各组按120mg/(kg·d)皮下注射D-半乳糖,连续6周,建立亚急性衰老模型。造模成功后,正常对照组和模型对照组灌胃生理盐水,其余各组灌胃相应浓度的药液。连续灌胃21天后,计算小鼠脾脏指数、胸腺指数;采用ELISA法测定血清中IL-2的浓度。结果与正常对照组比较,模型对照组小鼠脾脏指数、胸腺指数均显著下降(P<0.01,P<0.01),血清中IL-2水平显著下降(P<0.01)。与模型对照组比较,维生素E组、乙醇提取液组和仿生酶解液组小鼠脾脏指数、胸腺指数均升高(P<0.05,P<0.05,P<0.01),而各组之间无显著性差异(P>0.05)。维生素E组和仿生酶解液组小鼠血清IL-2水平均高于模型对照组(P<0.05,P<0.05),乙醇提取液组血清中IL-2水平亦有一定程度的升高,但不具有统计学意义(P>0.05)。结论蛤蚧乙醇提取液和仿生酶解液对D-半乳糖诱导衰老小鼠机体免疫功能均有一定的调节作用,且仿生酶解液组优于乙醇提取液组。 Objective To explore the effects of different extract of Gekko gecko Linnaeus on immune function of aging model mice induced by D-galactose( D-gal). Methods 50 KM mice were randomly divided into 5 groups: normal control group,model control group,Vitamin E group,ethanolic extract group,bionic enzyme hydrolysate group. Normal control group mice were given subcutaneous injection of physiological saline,other groups were given subcutaneous injection of D-gal at 120 mg/( kg · d). Six weeks later,normal control group and model control group were administrated of physiological saline,other groups were respectively administrated corresponding concentration of different extract. After 21 days,the indexes of thymus and spleen were accounted,and the concentration of Interleukin-2( IL-2) in serum were determined by ELISA method. Results Compared with normal control group,the indexes of thymus and spleen in model control group were significantly decreased( P〈0. 01,P〈0. 01),and the concentration of IL-2 in serum were declined significantly( P〈0. 01). Compared with model control group,the indexes of thymus and spleen in Vitamin E group,ethanolic extract group and bionic enzyme hydrolysate group were increased significantly( P〈0. 05,P〈0. 05,P〈0. 01),but there was no significant difference among three groups( P〈0. 05). The level of IL-2 in Vitamin E group and bionic enzyme hydrolysate group were higher than that of model control group( P〈0. 05). The level of IL-2 in ethanolic extract group was increased than that of model control group but there was no significant difference( P〈0. 05). Conclusion Ethanolic extract and bionic enzyme hydrolysate of Gekko gecko Linnaeus have certain effect on immune function in the aging model mice induced by Dgal,and the bionic enzyme hydrolysate is shown to be superior to ethanolic extract.
