双酚A对SD大鼠睾酮含量及P450scc和3β-HSD蛋白表达的影响 被引量:2

摘要 目的研究双酚A对雄性大鼠睾丸间质细胞内睾酮含量及睾酮合成关键酶P450scc和3β-HSD表达的影响,探讨双酚A对睾酮的作用机制。方法选择成年雄性SD大鼠48只,随机分为4组:对照组及染毒低剂量组(2 mg·kg-1·d-1)、中剂量组(20 mg·kg-1·d-1)、高剂量组(200 mg·kg-1·d-1)。连续灌胃8周。测定不同剂量组大鼠的血清睾酮含量,免疫组化方法检测睾丸间质细胞上P450scc和3β-HSD蛋白的表达。结果染毒中剂量组和高剂量组睾酮含量明显低于对照组(P<0.05),并有随染毒剂量增加而睾酮含量降低趋势;免疫组化图像分析结果显示:P450scc和3βΗSD在各染毒组平均吸光度值均数皆小于对照组,中剂量组和高剂量组与对照组相比差异有统计学意义(P<0.05)。结论双酚A可以抑制睾丸间质细胞合成睾酮,并抑制睾酮合成关键酶P450scc和3βHSD在睾丸间质细胞的表达。 Objective To observe the effects of bisphenol A( BPA) on the content of testosterone in testis of male rats and on the expressions of P450 scc and 3βhydroxysteroid dehydrogenase( 3β-HSD),and to explore its action mechanism.Methods Forty-eight adult male SD rats were randomly divided into 4 groups: control group,low dose group( 2 mg·kg-1·d-1),middle dose group( 20 mg · kg-1· d-1) and high dose group( 200 mg · kg-1· d-1),with continuous intragastric administration for 8 weeks. The serum levels of testosterone were measured in different dose groups,moreover the expression levels of P450 scc and 3β-HSD protein in testicular interstitial cell were detected by immunohistochemistry. Results The testosterone levels in middle dose group and high dose group were significantly lower than those in control group( P〈0. 05),meanwhile,the testosterone levels were gradually decreased with the increasing of the BPA dose. The immunohistochemical image analysis showed that the average absorbance values of P450 scc and 3β-HSD in the three different dose groups were obviously lower than those in control group,moreover,which in middle dose group and high dose group were significantly lower than those in control group( P〈0. 05). Conclusion The bisphenol A can inhibit the synthesis of testosterone and the expressions of the P450 scc and 3β-HSD in testicular interstitial cells of SD rats.
