摘要
目的:探讨特应性皮炎(AD)外周血T淋巴细胞亚群及其相关细胞因子与中医证型的内在联系。方法:AD患者风湿蕴肤证20例,血虚风燥证34例,健康对照组20例,采用流式细胞技术及ELISA法检测不同证型AD患者和对照组外周血中CD3^+、CD4^+、CD8^+、Treg及相关细胞因子的表达。结果:AD患者CD4^+、IL-4、IL-17表达均高于对照组,风湿蕴肤证及血虚风燥证组CD4^+表达差异无统计学意义;血虚风燥证CD8^+、IL-10表达高于风湿蕴肤证组及对照组,风湿蕴肤证及血虚风燥证组Treg表达均低于对照组,风湿蕴肤证Treg及IL-12表达低于血虚风燥证组,IL-4及IL-17表达高于血虚风燥证组。结论:可根据IL-4、IL-12、IL-17、IL-10水平及CD4^+、CD8^+、Treg数量初步判断病情阶段及中医辨证分型。
Objective: To explore the correlation between TCM syndrome and T lymphocyte subpopulation and its related cytokines. Methods: There were 20 patients in the syndrome of wind-damp stagnating in skin( WSS) group and 34 in the syndrome of wind and dryness due to blood deficiency( WDB) group. 20 patients were in the healthy control group. The expression of CD3^+,CD4^+,CD8^+,and Treg in peripheral blood were measured by flow cytometry( FACS). IL-4,IL-12,IL-17 and IL-10 in peripheral blood were detected by enzyme-linked immunosorbent assay( ELISA). Results: The expression of CD4^+,IL-4,IL-17 of AD group was significant higher than control group. There was no significant difference of the expression of CD4^+ in the WSS group and WDB group. The expression of CD8^+ and IL-10 of the WDB group was significant higher than the WSS group and control group. The expression of Treg in AD group was significant lower than control group. The expression of Treg and IL-12 of the WSS group was significant lower than the WDB group. The expression of IL-4 and IL-17 in WSS group was significant higher than The WDB group. Conclusion: The TCM syndrome differentiation of AD can be determined by the level of IL-4,IL-12,IL-17,IL-10,CD4^+,CD8^+ and Treg.
