摘要
目的探讨乌司他丁对大鼠肿瘤周围水肿的影响及其分子机制。方法采用大鼠原位胶质瘤模型,将大鼠随机分为2组:对照组和乌司他丁治疗组。于治疗1周后通过核磁共振成像检测肿瘤体积和水肿体积,蛋白印迹法检测细胞紧密连接蛋白Claudin-1的表达变化。结果乌司他丁治疗组肿瘤体积明显减小,水肿体积明显减小,细胞紧密连接蛋白Claudin-1的表达明显上升。结论乌司他丁通过上调Claudin-1的表达抑制肿瘤的生长和肿瘤周围水肿的发生,为其在临床的应用提供理论基础。
Objective To investigate the effect of Ulinastatin on peritumoral edema in rats and its molecular mechanism. Methods The rat model of orthotopic glioma was established, and the rats were randomly divided into two groups: the control group and Ulinastatin group. The tumor volume and edema volume were measured by magnetic resonance imaging at one week after the treatment. Western Blot was used to determine the expression change of cell tight junction protein Claudin-1. Results In the Ulinastatin treatment group, the tumor volume and edema volume significantly decreased and the expression of cell tight junction protein Claudin-1 significantly increased. Conclusion Ulinastatin inhibits tumor growth and peritumoral edema by up-regulating the expression of Claudin-1, which provides a theoretical basis for its clinical application.
