拔罐疗法对寒凝血瘀型家兔局部皮肤相关因子影响的实验研究 被引量:9

摘要 目的采用不同拔罐时间干预寒凝血瘀家兔模型,从家兔局部皮肤相关因子组织学角度探索拔罐疗法的部分生物学作用。方法采用冰冷刺激的方法复制寒凝血瘀家兔模型,选取大椎穴及其下方部位进行拔罐干预,检测拔罐局部皮肤的相关因子的含量变化和阳性表达。结果家兔血清中血管内皮生长因子(VEGF)含量测定:拔罐14 d组家兔血清VEGF含量与造模前相比差异具有统计学意义(P<0.05),而拔罐治疗14 d后VEGF含量上升,与造模后相比升高,差异具有统计学意义(P<0.05);家兔局部皮肤免疫组化切片的测定:拔罐治疗组(共3组)大椎穴拔罐处免疫切片VEGF、EGF和i NOS含量呈阳性表现;拔罐干预后寒凝血瘀型家兔皮肤SOD测定结果显示,拔罐组与空白组和模型组比较差异有统计学意义。结论通过研究局部皮肤的生长因子、炎性因子的蛋白表达,证实拔罐局部血管扩张,炎症反应减轻。推断拔罐疗法首先对局部皮肤产生良性物理刺激,改善微循环,通过影响生长因子、炎性因子等表达产生持续弱刺激,形成级联反应,进而影响整体微循环,产生拔罐疗法的"通络"效应。 Objective Different cuppingtime was used to intervene the rabbit model of cold,blood stasis and stasis,and some biological functions of cupping therapy were explored from the histology of local skin related factors in rabbits.Methods Model rabbits of congealing cold and blood stasis were copied with freeze stimulation,then selected dazhui point as the cupping treatment of parts,detected the content of cupping therapy and factors related to local skin changes and positive expression.Results The content of vascular endothelial growth factor(VEGF) in serum of rabbits: the serum VEGF content of rabbits in cupping 14 days group was significantly different from that before modeling(P〈0.05),but after 14 days of cupping treatment,the VEGF content increased,which increased significantly compared with that after modeling,and the difference was significant(P〈0.05).Rabbit skin immunohistochemistry determination: cupping therapy group(three groups) at Dazhui cupping section VEGF,EGF and i NOS immune showed positive content; cupping intervention after the cold coagulation and blood stasis rabbit skin SOD determination showed that the cupping group compared with the control group and model group,there was significant difference.Conclusion By studying the protein expression of growth factors and inflammatory factors in local skin,it is proved that the local vascular dilatation and inflammatory response are relieved.Cupping therapy first produces benign physical stimulation on the local skin,improves microcirculation,and produces continuous weak stimulation by affecting growth factors and inflammatory factors,resulting in cascade reaction,which affects the overall microcirculation and produces the "Tongluo" effect of cupping therapy.
