耳蜗干细胞移植联合中药干预对感音性耳聋治疗效果的影响

Effect of cochlear stem cell transplantation combined with traditional Chinese medicine on the treatment of sensorineural deafness

目的探讨耳蜗干细胞移植联合中药干预对感音性耳聋治疗效果的影响。方法随机将感音性耳聋豚鼠分成空白对照组,耳蜗干细胞移植组和干细胞移植+中药干预组,并将生理盐水、耳蜗干细胞悬液以及含有药物血清的耳蜗干细胞悬液分别向感音性耳聋豚鼠内耳耳蜗进行移植。在完成移植后分别于7 d,28 d,56 d对3组豚鼠进行免疫荧光观测实验以及ABR检测。结果耳蜗干细胞移植组和干细胞移植+中药干预组在进行干预后均可观测到MyosinⅦA阳性细胞及Nestin阳性细胞,与耳蜗干细胞移植组比较,干细胞移植+中药干预组的MyosinⅦA阳性细胞及Nestin阳性细胞数量显著升高,经比较差异具有统计学意义(P<0.05);在进行干预后,耳蜗干细胞移植组和干细胞移植+中药干预组ABR检测数值均出现下调,与耳蜗干细胞移植组比较,干细胞移植+中药干预组豚鼠的听力显著改善。结论耳蜗干细胞移植联合中药干预对感音性耳聋治疗效果明显,归芪地黄汤干预不仅可以明显提高耳蜗干细胞的存活率,还可以明显提高其分化为毛细胞的比例。

Objective To explore the effect of cochlear stem cell transplantation combined with traditional Chinese medicine on the treatment of sensorineural deafness.Methods Guinea pigs with sensorineural deafness were randomly divided into blank control group,cochlear stem cell transplantation group,stem cell transplantation plus traditional Chinese medicine intervention group,and normal saline,cochlear stem cell suspension and cochlear stem cell suspension containing drug serum were transplanted into the inner ear and cochlea of guinea pigs.At 7,28 and 56 days after treatment,all guinea pigs were detected by immunofluorescence observation experiment and ABR.Results Cochlea stem cell transplantation group and stem cell transplantation combined with traditional Chinese medicine in the intervention group after the intervention were observed Myosin Ⅶ A,Positive cells and Nestin positive cells,compared with cochlear stem cell transplantation group,stem cell transplantation combined with traditional Chinese medicine intervention group＇s VII A,the number of Myosin positive cells and Nestin positive cells were significantly increased,the difference was statistically significant（P〈0.05）.After intervention,the ABR values in the cochlear stem cell transplantation group and the stem cell transplantation plus Chinese medicine intervention group were all down regulated.Compared with the cochlear stem cell transplantation group,the hearing improvement in the stem cell transplantation plus Chinese medicine intervention group was significantly improved.Conclusion Cochlear stem cell transplantation combined with traditional Chinese medicine intervention is effective in the treatment of sensorineural hearing loss.The intervention of Guiqi Dihuang Decoction can not only significantly improve the survival rate of cochlear stem cells,but also significantly improve the proportion of differentiated cells to hair cells.
