白藜芦醇对大鼠肺缺血再灌注损伤的保护作用 被引量:2

The Protective Effects of Resveratrol on Lung Ischemia-reperfusion Injury in Rats

摘要 目的:探讨白藜芦醇(RSV)对大鼠肺缺血再灌注损伤(IRI)的保护作用及机制。方法:将30只SD大鼠随机均分为假手术组(Sham组)、IRI组和RSV组,IRI组和RSV组大鼠通过结扎大鼠右侧肺门建立大鼠IRI模型,Sham组大鼠仅做手术、不结扎右侧肺门,RSV组造模前30 min给予RSV腹腔注射、Sham和IRI组同时给予等量生理盐水腹腔注射;IRI组和RSV组大鼠于缺血再灌注6 h后(Sham组于相应时间)取动脉血检测二氧化碳分压(PaCO_2)和氧分压(PaO_2),同时取肺组织,利用物理法检测肺组织湿干质量之比(W/D),过碘酸雪夫染色(PAS)检测肺组织病理形态,TUNEL法检测肺脏细胞凋亡,Western blotting检测肺组织沉默信息调节因子1(SIRT1)、p53、乙酰化p53(Acetyl 53)、p21、Bax和BCL-2的表达。结果:与IRI组相比,RSV可降低肺组织W/D,上调PaO_2、下调PaCO_2,减轻肺组织病理损伤及细胞凋亡,显著增加肺组织SIRT1和BCL-2的表达,但明显减少Acetyl p53、p21及Bax表达,对p53表达无影响。结论:RSV可通过抗凋亡作用减轻肺IRI,其作用机制与调节SIRT1/p53信号通路相关。 Objective: To investigate the protective effect and mechanism of resveratrol on lung ische- miareperfusion injury in rats. Method: Thirty SD rats were randomly divided into sham operation group (Sham), ischemia reperfusion injury group (IRI) and resveratrol (RSV) group. The model of lung IRI was established By ligating the right hilum in IRI group and RSV group. RSV group was given intraperitoneal injection of resveratrol at 30 rain before operation, Sham and [RI groups were injected intraperitoneally with normal saline at the same time. Arterial blood was taken to detect the partial pressure of carbon dioxide (PaCO2) and oxygen partial (PaO2) after 6 h of ischemia and reperfusion. The physical method was used to detect lung wet to dry ratio (W/D) ; PAS staining was used to detect the pathological morphology of lung tissue ; In addition, TUNEL method was used to detect the apopto- sis of lung cells and Western blotting was used to detect the expression of silent information regulator 1 (SIRT1), p53, aeetyl p53, p21, Bax and BCL-2. Results: Compared with IRI group, resveratrol could significantly increase PaO2 and the expression of SIRTI, BCL-2, but significantly reduced PaCO2, W/D ratio of lung tissue and the expression of Acetyl p53, p2l, Bax, and alleviated patholog- ical injury of lung tissue and apoptosis. In addition, no influence was detected on the expression of.p53. Conclusion:Resveratrol can attenuate lung ischemia-reperfusion injury by anti- apoptosis, anti its mechanism is related to regulating SIRT1/p53 signaling pathway.
