髓样分化因子88基因多态性和肺结核易感的相关性分析

摘要 目的分析髓样分化因子88(MyD88)-938C/A和1944 C/G两个基因多态性位点和肺结核病易感性之间的关系。方法收集2016年10月—2017年3月河北省胸科医院结核科住院的102例初治肺结核患者作为病例组,同时选取同期健康体检者115例为健康对照组。应用聚合酶链反应—限制性片段长度多态性(PCR-RFLP)技术对2组研究对象MyD88-938C/A和1944 C/G多态性进行基因分型,分析其位点多态性与结核病发生是否相关。结果病例组和健康对照组MyD88-938C/A和1944 C/G位点基因型分析结果符合Hardy-Weinberg平衡定律。MyD88-938C/A位点存在3种基因型CC、CA和AA,2组3种基因型的分布频率及C、A等位基因分布频率差异有统计学意义(χ~2=13.24、11.64,P<0.01)。MyD88 1944 C/G位点有CC、CG和GG 3种基因型的分布,2组3种基因型的分布频率及C、G等位基因分布频率差异均无统计学意义(χ~2=1.28、1.11,P>0.05)。结论 MyD88-938C/A可能是肺结核发病的易感基因,而MyD88 1944 C/G基因与肺结核感染的易感性可能无关。 Objective Explore the relationship of MyD88-938C/A and 1944C/G gene polymorphisms with suscepti- bility to tuberculosis. Methods sputum smear positive of 102 initial treatment pulmonary tuberculosis patients were analyzed ( case group) in chest hospital of Hebei province from October 2016 to March 2017. MyD88-938C,/A and 1944C/G of 102 TB patients and 115 healthy controls were genotyped by polymerase chain reaetion-restrietion fragment length potymorphism (PCR- RFLP). Analysis the correlation between gene polymorphism of them and susceptibility to tuberculosis by X2 test. Results The geuotype analysis of MyD88-938C/A and 194-4C/G in the tuberculosis group and the control group accord with the law of Hardy-Weinberg equilibrium. The difference of CC, CA and AA genotype frequency of MyD88~938C/A were statistically sig- nificant between the case group and the control group by x2test and the difference of C,A allele frequency distribution in two groups were statistically significant( X2 = 13.24,11.64,P 〈 0.01 ). The difference of CC, C G and GG gnnotype frequency of MyD88 1944C/G were not statistically significant between the case group and the control group by xatest; while the difference of C, G allele frequency distribution in two groups were not statistically significant( X2=1.28,1.11, P 〉 0.05 ). Conclusion MyD88-938C/A may be the susceptibility gene of tuberculosis and MyD88 1944C/G may not be related to the susceptibility of tuberculosis infection.
