脂肪因子Chemerin促进RF/6A细胞增殖、迁移和管腔形成 被引量:1

Research on the Effect of Chemerin on RF/6A Cell Proliferation, Migration and Tube Formation

摘要 目的:研究Chemerin对体外培养的恒河猴脉络膜视网膜血管内皮细胞(RF/6A细胞)增殖、迁移和管腔形成的影响。方法:实验研究。取体外培养的生长良好的RF/6A细胞用于实验,将细胞随机分为4组,分别是对照组、5 ng/ml浓度Chemerin组、10 ng/ml浓度Chemerin组、20 ng/ml浓度Chemerin组。在RPMI-1640 培养液中进行培养24、48 h后,采用MTT法检测细胞增殖,划痕法检测细胞迁移,基质胶法检测细胞管腔形成。多组间比较采用单因素方差分析。结果:MTT检测结果提示,同对照组比较,10、20 ng/ml浓度Chemerin能够显著促进细胞增殖,组间差异均有统计学意义(P <0.05),5 ng/ml浓度组与对照组差异无统计学意义;划痕法检测细胞迁移结果提示,同对照组比较,3 种Chemerin浓度均能够显著促进细胞迁移,呈剂量依赖性,组间差异均有统计学意义(P <0.05);基质胶法检测细胞管腔形成结果提示,同对照组比较,3 种Chemerin浓度均能够显著促进细胞管腔形成,呈剂量依赖性,组间差异均有统计学意义(P <0.05)。结论:脂肪因子Chemerin能够促进RF/6A细胞增殖、迁移和管腔形成,提示Chemerin在视网膜新生血管形成中起重要作用。 Objective: To investigate the effects of chemerin on the proliferation, migration and tube formation of choroid retinal vascular endothelial cells (RF/6A) in the rhesus monkey in vitro. Methods: In this experimental study, RF/6A cells were randomly divided into 4 groups: a control group, and 3 chemerin groups treated with concentrations of 5, 10 or 20 ng/ml. The cells were cultured in a RPMI-1640 medium.After 24 h and 48 h in the culture, the MTT method was used to detect RF/6A cell proliferation. Cellmigration was detected by scratch test, and cell tube formation was detected by matrigel. A one-way analysis of variance was used to compare the data of the multiple groups. Results: MTT results showed that 10 ng/ml and 20 ng/ml concentrations of chemerin significantly promoted cell proliferation compared to the control group (P 〈 0.05). There was no significant difference between the 5 ng/ml concentration group and the control group. Cell migration results showed that the three different concentrations of chemerin significantly promoted cell migration in a dose-dependent manner. The difference between groups was statistically significant (P 〈 0.05). The three different concentrations of chemerin significantly promoted cell tube formation in a dose-dependent manner (P 〈 0.05) in the treated groups compared to the control group. Conclusions: Chemerin can promote the proliferation, migration and tube formation of RF/6A cells, suggesting that chemerin may play an important role in retinal neovascularization.
