摘要
目的探讨花仙宫血胶囊对功能失调性子宫出血大鼠模型子宫的保护作用。方法采用米非司酮联合米索前列醇建立大鼠功能失调性子宫出血模型。随机分为模型组、宫血宁胶囊(0.13 g/kg)组以及花仙宫血胶囊0.23、0.46、0.92 g/kg组,每组各10只。对照组和模型组ig等体积蒸馏水,其余各组于受孕第5天开始给药,每天给药1次,体积均为10 mL/kg,连续给药10 d。于末次给药后,次日处死大鼠,迅速摘取子宫、卵巢,计算脏器系数。采用固相夹心法酶联免疫吸附实验进行检测内膜血管内皮生长因子(VEGF)、基质金属蛋白酶-1(MMP-1)表达水平;观察HE染色后子宫组织形态学的变化。结果花仙宫血胶囊0.23、0.46、0.92 g/kg对子宫和卵巢指数均有增加趋势,但无显著性差异。花仙宫血胶囊0.46、0.92 g/kg明显升高子宫组织的VEGF表达水平(P<0.01),0.23、0.46、0.92 g/kg明显降低子宫组织的MMP-1表达水平(P<0.01)。花仙宫血胶囊对间质纤维结缔组织增生和炎细胞浸润有所改善,甚至可见新生血管。结论花仙宫血胶囊对功能失调性子宫出血大鼠具有显著的子宫保护作用,其作用机制与促进性器官的生长和成熟,调节子宫内膜VEGF、MMP-1表达水平有关。
Objective To discuss the protective effect of Huaxian Gongxue Capsules on uterus in dysfunctional uterine bleeding rats. Methods Dysfunctional uterine bleeding rat models were induced by misoprostol and mifepristone. Rats were randomly divided into model group, Gongxuening Capsules(0.13 g/kg) group, and Huaxian Gongxue Capsules(0.23, 0.46, and 0.92 g/kg) groups, and each group had 10 rats. Rats in the control and model group were given the same volume of water. The rest groups were ig administrated with the volume of 10 mL/kg from 5 th day of pregnancy, first daily, and were treated for 10 d. After the last administration, rats were killed on the next day, and the uterus and ovaries were quickly removed, and the organ coefficient was calculated. The expressions of VEGF and MMP-1 in endometrium were detected by solid phase sandwich ELISA. Morphologic changes of uterus were observed after HE staining. Results Huaxian Gongxue Capsules could increase the index of uterus and ovary, but there was no significant difference. Huaxian Gongxue Capsules(0.46 and 0.92 g/kg) could significantly increase the levels of VEGF(P〈0.01) and Huaxian Gongxue Capsules(0.23, 0.46, and 0.92 g/kg) could decrease the levels of MMP-1(P〈0.01). The proliferation of connective tissue and inflammatory cell infiltration in the interstitial fiber in Huaxian Gongxue Capsules groups were improved, and even the new blood vessels were visible. Conclusion Huaxian Gongxue Capsules have remarkable protection on uterus in dysfunctional uterine bleeding rats, and the mechanism is related to growth and maturation of sexual organs and the regulation of expression of VEGF and MMP-1 in the endometrium.
