摘要
目的研究雷公藤红素对P物质(susbstance p,SP)介导的肥大细胞活化的影响,方法应用CCK8检测雷公藤红素对小鼠P815肥大细胞的毒性作用,筛选出最佳的浓度和时间后将细胞分为细胞空白组,SP模型组,低、中、高浓度雷公藤红素组,雷公藤红素预处理细胞后,再采用SP刺激肥大细胞,致使其活化。采用ELISA检测组胺的释放量,RT-PCR检测NK-1R mRNA表达情况。结果雷公藤红素对细胞的毒性作用呈浓度时间依赖性,随着浓度和时间的增加毒性越大;与模型组相比,组胺的释放量随浓度的增加而减少,肥大细胞表面NK-1R mRNA表达下降(P<0.05),差异有统计学意义。结论雷公藤红素能抑制小鼠P815肥大细胞组胺的释放,可能跟NK-1R的表达有关。
Objective To study the effect of Celastrol on the activation of mast cells mediated by susbstance P (SP). Methods The toxic effects of Celastrol on cells were detected by CCKS. After screening the best concentration and time, The cells were divided into cell blank group, SP model group, low, medium and high concentration of Celastrol group and pretreat- ment cells with celastrol, then stimulated with SP, ELISA detected the release of histamine, The expression of NK-1R mRNA was detected by RT-PCR. Results The toxic effect of Celastrol on ceils was time-dependent, and the toxicity was added with the increase of concentration and time. Compared with the model group, the release of histamine decreased with the increase of concentration. The expression of NK-1R mRNA on the Mast Cell surface were decreased (P〈0. 05), which was statistically significant. Conclusion Celastrol can suppresses the release of mast cells histamine in mouse P815, which is related with the expression of NK-1R.
