妊娠期糖尿病产妇局麻药神经毒性损伤的机制探索

摘要 目的:通过细胞试验对妊娠期糖尿病产妇局麻药神经毒性损伤的机制进行探索。方法:研究使用人未分化SH-SY5Y神经母细胞瘤细胞培养,细胞分为4组,不同时间点观察SH-SY5H细胞活力、凋亡率等,以评价妊娠期糖尿病产妇局麻药神经毒性情况,统计分析使用SPSS20.0分析。结果:研究发现各组细胞除了具有形态学上的不同之外,对照组细胞4个不同的时间点细胞活力未见明显改变;A组细胞在4个时间点的细胞活力变动也不大;B组细胞及C组细胞可见随着孵育进程细胞活力逐渐减低,特别是B组细胞活力减退显著;B、C组细胞在孵育1h、孵育6h以及孵育24h时间点的细胞活力均显著低于开始时刻水平。对照组和A组细胞凋亡未见明显改变;B组细胞以及C组细胞可见随着孵育进程细胞凋亡率逐渐增加,特别是B组细胞最为显著;B、C组细胞在孵育1h、孵育6h以及孵育24h时间点的细胞凋亡率均显著高于开始时刻水平。结论:AKT/ERK信号通路失衡状态及伴随的离子通道状态改变与妊娠高糖状况下局麻出现神经毒性反应存在关联。 Objective:To explore the mechanism of neurotoxic injury of local anesthetic in pregnant women with gestational diabetes by cell test.Methods:The cultured human undifferentiated SH-SY5 Yneuroblastoma cells were cultured and divided into 4 groups.The viability and apoptosis rate of SH-SY5 H cells were observed at different time points.to evaluate the neurotoxicity of local anesthetic in pregnant women with gestational diabetes.Statistical analysis was analyzed using SPSS 20.0.Results:The results showed that in addition to morphological differences,the cells in control group showed no significant changes in cell viability at four different time points.The changes of cell viability in the mesanglion cells at four different time points were not significant;Cells in group B and cells in group C showed a gradual decrease in cell viability during the incubation period,especially in group B;the viability of cells in group B and group C at 1 hour of incubation,12 hours of incubation,and 24 hours of incubation were significantly lower than those at the start moment.There was no significant change in the apoptosis of the control group and the group A;The apoptosis rate of the cells in the group B and group C was gradually increased with the progress of the incubation,especially the cells in the group B were the most obvious;In the cells in the group B and group C,the apoptosis rate at 1 hour of incubation,12 hours of incubation,and 24 hours of incubation were significantly higher than those at the initial time.Conclusion:The imbalance of AKT/ERK signaling pathway and the accompanying changes of ion channel status may be related to the neurotoxic reaction of local anesthesia in gestational hyperglycemia.
