补脾强力复方对EAMG大鼠Th1/Th2/Th17细胞因子的影响 被引量:3

Effect of potent compoundspleen-tonifying prescription on Th1/Th2/Th17 cytokines in a rat model of experimental autoimmune myasthenia gravis

摘要 目的:研究补脾强力复方对实验性自身免疫性重症肌无力(EAMG)大鼠血清Th1/Th2/Th17细胞因子的影响,探讨其可能的作用机制。方法:选择健康SPF级雌性Lewis大鼠,分为正常组、模型组、醋酸泼尼松组及补脾强力复方高、中、低剂量组,采用免疫诱导注射鼠源性AchR-α亚基97-116肽段方法制备大鼠EAMG模型。ELISA法检测各组大鼠血清细胞因子IFN-γ、TGF-β、TNF-α、IL-2、IL-4、IL-17水平。结果:与正常组比较,模型组大鼠血清中IFN-γ、TNF-α、IL-2、IL-4、IL-17水平均明显升高,TGF-β水平明显降低,差异均有统计学意义(P<0.05或P<0.01);与模型组比较,补脾强力复方高、中、低剂量组大鼠血清IFN-γ、TNF-α、IL-2、IL-4、IL-17水平明显降低,而TGF-β水平明显升高,差异均有统计学意义(P<0.01或P<0.05);醋酸泼尼松组大鼠血清TNF-α、IL-2、IL-4、IL-17水平较模型组明显降低,差异均有统计学意义(P<0.01)。结论:补脾强力复方能有效降低EAMG大鼠IFN-γ、TNF-α、IL-2、IL-4、IL-17水平,升高TGF-β水平,从而纠正EAMG大鼠Th1/Th2/Th17细胞因子失衡,这可能是其治疗EAMG的作用机制之一。 Objective: To investigate the effect of potent compoundspleen-tonifying prescription on Th1/Th2/Th17 cytokines in a rat model of experimental autoimmune myasthenia gravis( EAMG) and possible mechanism of action. Methods: Healthy specific pathogen-freefemale Lewis rats were divided into normal group,model group,prednisone acetate group,and high-,medium-,and low-dose potent compoundspleen-tonifying prescription groups. A rat model of EAMG was established by immune induction with mouse-derived AchR-α subunit 97-116 peptide. ELISA was used to measure the serum levels of interferon gamma( IFN-γ),transforming growth factor-β( TGF-β),tumor necrosis factor-α( TNF-α),interleukin-2( IL-2),interleukin-4( IL-4),and interleukin-17( IL-17). Results: Compared with the normal group,the model group had significant increases in the serum levels of IFN-γ,TNF-α,IL-2,IL-4,and IL-17 and a significant reduction in the serum level of TGF-β( P〈0. 05 or P〈0. 01). Compared with the model group,the high-,medium-,and low-dose potent compoundspleen-tonifying prescription groups had significant reductions in the serum levels of IFN-γ,TNF-α,IL-2,IL-4,and IL-17 and a significant increase in the serum level of TGF-β( P〈0. 01 or P〈0. 05). Compared with the model group,the prednisone acetate group had significant reductions in the serum levels of TNF-α,IL-2,IL-4,and IL-17( P〈0. 01). Conclusion: Potent compoundspleen-tonifying prescriptioncan effectively reduce the serum levels of IFN-γ,TNF-α,IL-2,IL-4,and IL-17,increase the level of TGF-β,and thus correct the imbalance of Th1/Th2/Th17 cytokines in EAMG rats,which may be the mechanism of action of this prescription in the treatment of EAMG.
