薏苡仁油对喉癌细胞侵袭迁移能力的影响

The effect of coix seed oil on migration and invasion of laryngeal cancer

目的研究薏苡仁油对喉癌细胞株Hep-2侵袭和迁移能力的影响。方法用含不同浓度(0、25、50、100、200、400、800、1 600μM)薏苡仁油的培养基分别培养人喉癌细胞株Hep-2 24、48、72、96 h,采用CCK8法观察薏苡仁油对Hep-2细胞的抑制率。采用细胞划痕实验和Transwell侵袭实验检测细胞的迁移、侵袭能力;Western blot检测侵袭迁移相关蛋白的表达情况;采用SPSS 17.0软件对数据进行统计学处理,以P<0.05为差异具有统计学意义。结果薏苡仁油抑制喉癌细胞株Hep-2的增殖,其抑制效应呈剂量-时间依赖性;薏苡仁油对喉癌细胞株Hep-2的半数抑制浓度(IC50)为:165.2~650.2μM;经薏苡仁油作用后Hep-2细胞的侵袭和迁移能力降低,差异具有统计学意义(P<0.05);薏苡仁油(100,200μM)作用后,与对照组相比,Vimentin、Slug、p ERK1/2的表达下调;E-cad的表达升高;ERK1/2的表达不变。结论薏苡仁油可抑制喉癌细胞株Hep-2的侵袭和迁移能力。

Objective To investigate the effect of coix seed oil on migration and invasion of laryngeal cancer. Methods Hep-2 laryngeal cancer cells were treated with different concentrations of OA(0, 25, 50, 100, 200, 400, 800 and 1 600 μM) for 24, 48, 72, 96 h. The cell inhibition rate was evaluated using a modified Cell Counting Kit-8(CCK8) assay. Wound healing assay and transwell assay were performed to analyze the effect of coix seed oil on invasion and migration in Hep-2 cells. The expression levels of proteins E-cad, Vimentin, Slug, ERK1/2, p ERK1/2 were detected by Western blotting. All statistical analyses were performed using the Statistical Package for the Social Sciences(SPSS, Chicago, IL), Version 17.0. The value P<0.05 was considered statistically signifcant. Results Coix seed oil inhibits the proliferation of Hep-2 cells in vitro. The anti-proliferation effect of coix seed oil on Hep-2 cells was portrayed in a dose-and time-dependent manner. The IC50 values of coix seed oil were 165.2-650.2μM. Coix seed oil inhibited the invasive and migrative ability on Hep-2 cells(P<0.05). The protein level of Vimentin, Slug, p ERK1/2 were decreased with the increasing concentrations of coix seed oil. The expression of E-cad was increased with the increasing concentrations of coix seed oil. ERK1/2 was not obviously changed in Hep-2 cells afer treatment with coix seed oil. Conclusion Coix seed oil inhibited the migration and invasion of Hep-2 laryngeal cancer cells.
