煤尘诱导巨噬细胞释放细胞因子和氧化损伤水平研究 被引量:1

摘要 目的探讨煤尘诱导人肺泡巨噬细胞释放细胞因子的影响。方法通过佛波酯(PMA)刺激THP-1细胞诱导其分化为人肺泡巨噬细胞,随机分为对照组,2 000μg/ml组(高剂量组),1 000μg/ml组(中剂量组)和500μg/ml组(低剂量组),煤尘染毒8 h,测定IL-lβ、IL-6、TNF-α。结果不同剂量的媒尘均可诱导巨噬细胞释放IL-lβ、IL-6、TNF-α,其中高剂量组的细胞因子水平升高最为明显,存在剂量反应关系。结论 IL-lβ、IL-6、TNF-α可能在煤工尘肺的纤维化过程中发挥重要作用。 Objective To investigate the effect of coal dust on the release of cytokines in human alveolar macrophages. Methods THP-1 cells were stimulated by phorbol ester(PMA) to induce their differentiation into human alveolar macrophages, and were randomly divided into 4 groups includeing one control group and 3 exposure groups(low-dose group, middle dose group and high dose group). They received different dose of coal dust by gavage for 8 hours continuously, and cytokines IL-lβ, IL-6, TNF-αwere measured. Results IL-lβ, IL-6, TNF-α were higher than that in contrast groups(P<0.05), and the cytokine level were the highest in high dose group(P<0.05), and there was dose response relationship. Conclusion IL-lβ, IL-6, TNF-αmay play an important role in the process of fibrosis of coal workers' pneumoconiosis.
