响应面法优化盐碱土中所得一株细菌的培养条件 被引量:7

Optimization of culture conditions of a bacterial strain from saline alkali soil by response surface methodology

摘要 以盐渍化土壤中分离得到的一株节杆菌(Arthrobacter sp.)HX-6为研究对象,在单因素试验的基础上,通过Box-Behnken试验设计和响应面分析法,考察培养基中不同p H、培养温度和培养基中Na Cl质量分数对菌株HX-6生长的影响,探索菌株HX-6最适培养条件。结果表明,3个因素对菌株HX-6的生长影响均显著,从而获得菌株HX-6的最适培养条件为p H值7.0,培养温度34℃,培养基中Na Cl含量0.5%;在此条件下培养11 h,菌株HX-6菌悬液在波长600 nm处的吸光度值为0.43,表示菌株生长良好。 Based on the single factor experiments, the culture conditions of strain Arthrobacter sp. HX-6 from saline alkali soil were optimize by Box-Benhnken experimental design and response surface analysis method. The effects of pH, culture temperature and sodium chloride concentration on the growth of strain HX-6 were investigated. Results showed that three factors had all significant effect on the grouth of strain HX-6 growth, and the optimum culture conditions were pH 7.0, temperature 34 ^ and sodium chloride concentration 0.5%. Under these conditions, the absorbance value of bacterial suspension of strain HX-6 at 600 nm was 0.43 after cultivation for 11 h, which indicated that the strain HX-6 grew well.
