摘要
建立超声浸提-高效液相色谱法快速测定刺三加叶中绿原酸含量的方法。选择ZORBAX SB-C_(18)色谱柱(4.6 mm×250 mm,5μm),以甲醇-0.2%磷酸水溶液(15∶85,V/V)为流动相,检测波长为327 nm,流速1.0 m L/min,柱温30℃。结果表明,绿原酸在0.08~4.16μg/m L质量浓度范围内与峰面积呈现良好的线性关系(相关系数R^2=0.999 6),检测限为0.007μg/m L,平均加样回收率为99.84%,相对标准偏差(RSD)为1.53%。该方法操作简便快捷、灵敏,结果准确,且具有良好的稳定性、重复性和准确性,可用于刺三加叶中绿原酸的含量测定。
The rapid determination of chlorogenic acid content in Acanthopanan trifoliatus leaves was established by ultrasonic extraction and HPLC.Agilent ZORBAX SB-C_(18)(4.6 mm×250 mm, 5 μm) was chromatographic column, methanol 0.2% phosphoric acid(15∶85,V/V) were mobile phase with flow rate of 1.0 ml/min, the determine wavelength was 327 nm and the column temperature was 30 ℃. The results showed that there was a good linear relationship between chlorogenic acid in the range of 0.08-4.16 μg/ml and peak area(R^2=0.999 6). The limit of detection(LOD) was 0.007 μg/ml,the average adding standard recovery rate was 99.84%, the relative standard deviation(RSD) was 1.53%. The method was simple, rapid, sensitive and accurate with good stability, repeatability and accuracy. It could be used for the determination of chlorogenic acid in A. trifoliates leaves.
