健脾疏肺食疗方对肺脾两虚证支气管哮喘模型大鼠白细胞介素4和干扰素γ的影响 被引量:6

摘要 目的观察健脾疏肺食疗方对肺脾两虚证支气管哮喘(以下简称哮喘)大鼠肺泡灌洗液(BALF)中白细胞介素4(IL-4)和干扰素γ(IFN-γ)含量的影响。方法将40只大鼠随机分为4组,分别为正常对照组、模型组、氨茶碱组、食疗组,每组各10只。除正常对照组外,模型组、氨茶碱组及食疗组大鼠以香烟喷雾、番泻叶浸液灌胃及卵清白蛋白磷酸缓冲液雾化吸入致肺脾两虚证哮喘模型。从造模开始第2 d,正常对照组、模型组及氨茶碱组大鼠予0.9%氯化钠注射液2 m L灌胃,其中在造模第21 d氨茶碱组将0.9%氯化钠注射液更换为氨茶碱液2 m L(生药含量为4.06 mg)持续灌胃。食疗组在造模开始第2 d予健脾疏肺食疗方药液2 m L(生药含量为2.87 g)持续灌胃。以上各组均持续灌胃至造模第30 d。比较4组大鼠实验第1 d及灌胃结束时体质量的差值,检测比较BALF中IL-4和IFN-γ含量。结果与正常对照组比较,模型组、氨茶碱组及食疗组大鼠体质量差值较小(P<0.05),大鼠体质量增加幅度有所降低;与模型组比较,氨茶碱组及食疗组大鼠体质量差值增加(P<0.05)。与正常对照组比较,模型组、氨茶碱组及食疗组大鼠BALF中的IL-4含量均升高(P<0.05),IFN-γ含量均降低(P<0.05);与模型组比较,氨茶碱组及食疗组IL-4含量均降低(P<0.05),IFN-γ含量均升高(P<0.05)。结论健脾疏肺食疗方能借助调节细胞因子IFN-γ以及IL-4的含量,抑制或阻断哮喘的早期发病过程,抗气道变态反应性炎症及免疫调节作用可能是其防治哮喘的机制之一。 Objective To observe the effects of Spleen-strengthening and Lung-unchoking diet prescription on the levels of interleukin-4( IL-4) and interferon-γ( IFN-γ) in bronchoalveolar lavage fluid( BALF) of bronchial asthma rat model with deficiency of lung-spleen syndrome. Methods 40 rats were randomly divided into four groups: normal control group,model group,aminophylline treatment group and diet group,10 rats in each group. Except the normal control group,the model group,aminophylline treatment group and diet group rats were given cigarette spray,senna extract,ovalbumin inhalation to induce asthma model with deficiency of lung-spleen syndrome. 2 days after the beginning of modeling,rats in the normal control group,model group and aminophylline treatment group were given 0. 9% sodium chloride injection 2 mL by gavage,twenty-first day 0. 9% sodium chloride injection was replaced by 2 mL aminophylline( crude drug content was 4. 06 mg) continued by gavage at twenty-first day. On the second day after the beginning of modeling,the diet group was given 2 mL of Spleen-strengthening and Lung-unchoking diet prescription( crude drug content was 2. 87 g) continued by gavage. The above groups were continued gavage until 30 th d. modeling. The levels of IL-4 and IFN-γ of BALF of were compared in four groups,and the difference of body mass of rat between 1 th d of experiment and the end of experiment were compared in fourgroups. Results Compared with the normal control group,the difference of body mass in model group,aminophylline treatment group and the diet group was smaller( P < 0. 05),and the increase range of body mass of rats reduced. Compared with model group, the difference of body mass inaminophylline group and diet group increased( P < 0. 05). Compared with normal control group,the contents of IL-4 of BALF in aminophylline group and diet group increased,and the content of IFN-γdecreased( P < 0. 05). Compared with the model group,the content of IL-4 in diet group and aminophylline group decreased,and the content of IFN-γincreased( P < 0. 05). Conclusion Spleen-strengthening and Lung-unchoking diet prescription can inhibit or block the early onset of asthma by regulating the levels of cytokines IFN-γand IL-4,and it has the function of resisting airway allergic inflammation and immune modulation,which may be one of the mechanisms of diet therapy to prevent and cure asthma.
