摘要
通过单因素实验,结合改进的Dische比色法测定岩藻糖含量,探究热水浸提海带中岩藻聚糖的最佳提取工艺。并对最佳条件下提取的产物进行初级纯化后,用红外光谱进行基本结构分析。结果表明,在液料比40∶1、提取温度90℃和提取时间4 h的最佳提取条件下所得的岩藻聚糖提取率最高,达10.61%(多糖中岩藻糖含量为20.94%),基本理化性质分析及红外光谱图表明所提取的多糖含有岩藻糖、糖醛酸及硫酸基团,并且端基碳构型可能为α-型吡喃糖。
In order to improve the yield of fucoidan,hot water extraction process of fucoidan from Laminaria japonica was optimized by mono-factorial experiment and fucose content,the content of L-fucose was determined by improved Dische colorimetry. The structure of polysaccharide was analyzed by infrared spectrum after primary purification of the product under the optimum conditions. The optimum process parameters for hot water extraction process were as follows:extraction liquid-material ratio was 40∶1,extraction temperature was 90 ℃,extraction time was 4 h. Under the above conditions,the highese yield of fucoidan was obtained which was up to 10. 61%( fucose content was up to 20. 94% in polysaccharide). Basic physical and chemical properties analysis and IR spectrum proved that the extracted polysaccharides contained fucose,uronic acid,sulfate group,and the configuration of glycoside bond of crude fucoidan was presumably α-pyranose.
