摘要
采用双波长分光光度法对西藏秦艽花中总黄酮测定进行实验,将从西藏秦艽花中提取出的异荭草苷定为对照品,选择406 nm与435 nm分别为该方法的测定波长与参比波长,通过AlCl_3显色法,得到西藏秦艽花药材的总黄酮含量为2.20%。分别以西藏秦艽花总黄酮提取时所用的提取溶剂浓度、提取时间和液固比为对象进行考察研究,再由正交试验表对总黄酮的提取进行实验设计,结果得到的西藏秦艽花总黄酮提取时所用的最佳条件是乙醇溶液浓度70%,超声提取时间40 min,液固比1∶25 g/mL。
The content of total flavonoids in flower of Gentiana tibetica was determined by double-wavelength spectrophotometry with AlCl_3 as chromogenic agent,choosing 406 nm as the detection wavelength and 435 nm as the reference wavelength with isoorientin as index. The total flavonoids content of the flower of Gentiana tibetica was 2. 20%.The optimum conditions of extraction from orthogonal experiment were 70% of ethanol concentration,40 min of ultrasonic extraction time and 1∶25 g/mL of liquid-solid ratio.
