摘要
用高效液相色谱法(HPLC)同时测定野核桃、胡萝卜两种食品中的β-胡萝卜素含量。采用的色谱条件为:Symmetry C18色谱柱(4.6 mm×250 mm,5μm),以60%乙腈、20%乙酸乙酯、20%甲醇混合溶液为流动相,在波长450 nm,柱温25℃流速1 m L/min;进样量10μL条件进行试验。在优化实验条件下,测得野核桃中含β-胡萝卜素为6.5μg/g,胡萝卜中含β-胡萝卜素为358.15μg/g。方法简便、快速,适合高效液相色谱法测定植物样品中β-胡萝卜含量。
The contents of β-carotene in wild walnut and carrot were simultaneously determined by HPLC. The chromatographic conditions were as follows: symmetry C18 column(4. 6 mm×250 mm,5 μm) was used with mobile phase of a mixture of 60% acetonitrile,20% methanol at a wavelength of 450 nm,and a column temperature of 25 ℃ with1 m L/min as the flow rate,injection volume of 10 μL. Under the optimized experimental conditions,the β-carotene content was obtained in wild walnut containing 6. 5 μg/g and in carrots containing 358. 15 μg/g for real samples. The method was simple,rapid and suitable for the determination of β-carrot content in plant samples by high performance liquid chromatography.
