Effect of berberine on proliferation and apoptosis of human ovarian cancer cells

摘要 目的探讨黄连素对人卵巢癌细胞株OVCAR3增殖和凋亡的影响,为卵巢瘤的治疗提供实验依据。方法采用不同浓度的黄连素处理人卵巢癌细胞株OVCAR3,等体积培养基培养正常对照组细胞。CCK-8法观察黄连素对卵巢癌细胞增殖活性的影响;流式细胞仪检测黄连素处理后细胞凋亡情况;RT-PCR、Western Blot分别检测卵巢癌细胞中Caspase-3、Caspase-8mRNA及蛋白表达情况。结果黄连素抑制人卵巢癌细胞株OVCAR3的增殖且呈时间剂量依赖性,并促进细胞凋亡。RT-PCR、Western Blot结果显示:黄连素作用于OVCAR3细胞株后,细胞Caspase-3、Caspase-8 mRNA及蛋白表达升高,差异有统计学意义(P<0.05)。结论黄连素能够通过抑制人卵巢癌细胞株OVCAR3增殖、促进凋亡发挥抗肿瘤作用,可为临床卵巢癌治疗策略提供新的思路。 Objective To explore the effect of berberine on proliferation and apoptosis of human ovarian cancer cells,provide an experimental basis for treatment of ovarian cancer. Methods Different doses of berberine were used to treat human ovarian cancer OVCAR3 cell line,and isometric culture medium was used to cultivate normal cells. CCK-8 method was used to observe the effect of berberine on proliferation activity of human ovarian cancer cells. Flow cytometry was used to detect cell apoptosis after treated by berberine. RT-PCR and Western Blot were used to detect the expressions of Caspase-3 and Caspase-8 mRNA proteins in ovarian cancer cells. Results Berberine could effectively inhibit proliferation of human ovarian cancer OVCAR3 cell line in vitro,showing a time-and dose-dependent manner,which promoted cell apoptosis. The results of RT-PCR and Western Blot showed that the expressions of Caspase-3 and Caspase-8 mRNA and proteins in ovarian cancer cells increased significantly after treated by berberine,there were statistically significant differences( P 0. 05). Conclusion Berberine has anti-tumor effect via inhibiting proliferation and promoting apoptosis of human ovarian cancer OVCAR3 cell line,which provide new ideas for developing clinical treatment strategies of ovarian cancer.
