不同生长期野葛与粉葛的活性成分及体外抗氧化活性研究 被引量:19

Studies on the Bioactive Components and Antioxidant Activity of both Puerarialobata(Willd.)Ohwi and P.thomsonii Benth during Different Growing Periods

摘要 对比不同生长期野葛和粉葛的植物化学素及体外抗氧化活性,测定野葛和粉葛的异黄酮含量。采用HPLC方法测定葛根中葛根素、大豆苷、大豆苷元、染料木苷、染料木素、芒柄花黄素、鹰嘴豆芽素、异甘草素等8种异黄酮含量,研究两个品种葛根主要活性物质含量积累动态变化规律,并用DPPH、ABTS和FRAP 3种体外抗氧化方法评价生长期两种葛根的体外抗氧化性,以确定其适宜采收期。结果表明:随着生长期增加,野葛和粉葛中的异黄酮含量均大幅增加,尤其是野葛的总异黄酮含量增加了53%。生长期野葛和粉葛中各异黄酮含量均显著性积累增加(P<0.05)。体外抗氧化性研究表明:野葛的体外抗氧化能力均高于粉葛,且随着生长期的增加,体外抗氧化活性随之增强,在12月份异黄酮含量和抗氧化活性达到最高水平。由此确定12月份适宜采收葛根。 To compare the bioactive components as well as the antioxidant effects in vitro of both Puerarialobata(Willd.)Ohwi and P.thomsonii Benth during different growing periods,the total isoflavones of them were determinated.Besides,the major isoflavones including puerarin,daidzein,daidzein,genistin,genistein,formononetin,isoliquiritigenin and biochanin A were determinate quantificationally with a HPLC method to study the dynamic accumulation of the bioactive compositions during different growing periods,so as to determine the suitable harvest period.Then,the antioxidant activities of the two kinds of Radix puerariae were evaluated by DPPH,ABTS and FRAP method in vitro.The results showed that: as the growing period increased,the content of isoflavone in both Puerarialobata and P.thomsonii significantly increased(P0.05),especially the content of total isoflavones of Puerarialobata(Willd.) Ohwi rose 53%.As the Puerarialobata grew,the isoflavone content of two kinds of Radix puerariae significantly increased.The vitro antioxidant research showed that the antioxidant ability of Puerarialobata were higher than that of P.thomsonii.With the increase of growing period,the vitro antioxidant activity also increased,in December both the isoflavone content and antioxidant activity reached the highest level,so we determine December as the suitable harvesting period.
