Isolation and Purification of Retinol Palmitate Synthesized by Immobilized Lipase Transesterification

摘要 利用冷冻结晶和萃取的方法对由固定化脂肪酶催化维生素A醋酸酯和棕榈酸合成的维生素A棕榈酸酯进行分离纯化,获得纯度高的维生素A棕榈酸酯。分离纯化维生素A棕榈酸酯的工艺流程为:将反应液先在4℃放置20 h,然后在-20℃放置8 h,除去过量的棕榈酸。液-液萃取,纯化。以84%乙醇-水∶石油醚=3∶1(v/v)为萃取溶剂,萃取温度4℃,萃取级数4次,此时维生素A棕榈酸酯纯度达到98.3%,本工艺为维生素A棕榈酸酯的生产提供了技术支持。 The paper studied the isolation and purification of retinol palmitate synthesized by immobilized lipase with retinyl acetate and palmitic acid as substrates. Higher purity retinol palmitate was obtained using freezing crystallization and extraction. The technological processes were as follows: firstly, the reaction solution was placed at 4 ℃ for 20 h and then changed the temperature to-20 ℃ for another 8 h to get rid of the excessive palmitic acid. Then, the liquid-liquid extraction was done for further purification. The extraction solvent was 84% ethanol/distilled water : petroleum ether(3∶1), the extraction temperature was 4 ℃, the extraction cycle was 4. After extraction, the purity of retinol palmitate reached 98.3%. This work provided technical basis for the production of retinol palmitate.
