金铁锁毛状根皂苷的薄层色谱分析及抑菌活性 被引量:5

TLC Analysis of Psammosilene tunicoides Hairy Roots Saponins and Its Antibacterial Activity

摘要 为了评价金铁锁毛状根的药用价值,对毛状根中主要活性成分皂苷进行分析,以及对毛状根提取物的抑菌活性进行检测。采用硅胶色谱法对毛状根中主要活性成分皂苷进行分析;采用牛津杯法以大肠杆菌、金黄色葡萄球菌和藤黄八叠球菌作为指示菌对毛状根提取物进行抑菌活性检测。发现氯仿:乙酸乙酯:甲醇:水(15:40:22:10)的展开系统分离的皂苷谱带效果好;与野生金铁锁药材相比,毛状根组织中也含有7种相同皂苷,另外7种皂苷未检出,但毛状根中出现2种新皂苷,而且毛状根组织中有3种低极性皂苷含量明显高于金铁锁药材。金铁锁毛状根的乙醇提取物对三种指示菌都有较强的抑制效果,优于金铁锁野生药材的抑菌效果。研究结果表明,金铁锁毛状根组织新合成的低极性皂苷可能是毛状根抑菌性强于野生药材的原因之一,但需要进一步研究新皂苷的生物合成机理、生理学意义和药物活性等。本研究结果为现如今在金铁锁毛状根方面的研究填补了空白,也为其后续的深入研究做了铺垫,对金铁锁毛状根未来的进一步开发起到重要的引导作用。 In order to evaluate the medicinal value of P. tunicoides hairy roots, the main active ingredient saponins in the hairy roots was analyzed by using TLC method, and the antibacterial activity of the hairy roots was tested by using oxford cup method with E. coli, S. aureus and S. lutea as indicator bacterias. The results showed that the saponins bands could be well separated in the developing solvent of chloroform: ethyl acetate: methanol: water(15:40:22:10). Compared with wild P. tunicoides herbs, the same seven kinds of saponins were identified and other seven kinds of saponins undetected, however, two novel saponins were discovered in hairy root and the concentration of three kinds of low polar saponins were higher than P. tunicoides herbs. The ethanol extract of P.tunicoides hairy roots displayed stronger ability to inhibit indicator bacteria growth than that of wild P. tunicoides roots. The above data suggested that the newly synthesized low polarity saponins may be one of the reasons that hairy roots were more potent than wild herbs, it's necessary to further study for their mechanisms of biosynthesis,physiological significance and pharmaceutical activity and so on. The results of this study not only filled the gaps in the study of P. tunicoides hairy roots, but also for its follow-up study done paved the way and played animportant role in guiding on the future development of P. tunicoides hairy roots.
