摘要
以两种羊肚菌菌种为实验材料,通过研究比较菌丝生长进度来寻找最适羊肚菌生长的基础培养基,在最适培养基基础上通过正交试验探究不同碳、氮源、pH值和温度如何影响菌丝生长的特性,得到影响羊肚菌菌丝生长条件的碳源、氮源、pH和温度的最优组合,结果显示,最适合两种羊肚菌菌株生长的培养基分别为:葡萄糖30 g/L,NaNO_31.5 g/L,KH2PO41 g/L,Fe SO40.025 g/L,琼脂12 g/L和可溶性淀粉20g/L,NaNO_32 g/L,KH2PO41 g/L,Mg SO40.5 g/L,琼脂粉12 g/L。正交试验和方差分析结果显示,两种羊肚菌的氮源因素各水平菌丝生长速率差异显著,而碳源、pH和温度各水平菌丝生长速率差异不显著;羊肚菌菌"Y"的四个因素碳源、氮源、pH和温度的最优组合可以为葡萄糖、NH_3NO_3、pH 8.5、温度30℃和土豆、NH_3NO_3、pH 7.5、温度20℃,羊肚菌"B"的四个因素的最优组合为可溶性淀粉、尿素、pH 6.5、温度30℃。
In this research we took two Morehella esculenta strains as experimental materials, Through comparisons of mycelia growth rates to search the optimal culture medium of Morehella esculenta. On the basis of optimal medium, the impacts of different carbon sources and nitrogen sources, pH and temperature on mycelia growth characteristics were tested by orthogonal test. The results showed that the optimal medium for the growth of two Morehella esculenta strains were glucose 30 g/L, NaNO_31.5 g/L, KH2 PO41 g/L, Fe SO40.025 g/L, agar 12 g/L and soluble starch 20 g/L, NaNO_32 g/L, KH2 PO41 g/L, Mg SO40.5 g/L, agar 12 g/L, respectively. The orthogonal test and variance analysis showed there were significant differences in the growth rates of mycelial growth between the two sources of nitrogen, but the difference of the growth rates of carbon sources, pH and temperature were not significant. The optimal combination of Morehella esculenta "Y"four factors of carbon, nitrogen,pH and temperature may be glucose, NH_3NO_3, pH8.5, temperature 30℃ and potatoes, NH_3NO_3, pH 7.5, temperature 20℃, the optimal combination of Morehella esculenta"B"four factors was soluble starch, urea, pH 6.5, and temperature 30℃.
