皮肤瘢痕癌中Bcl-2蛋白的表达与癌细胞凋亡相关性的探讨

摘要 目的探讨瘢痕癌Bcl-2蛋白的表达与癌细胞凋亡相关性。方法以15例病理性瘢痕、20例皮肤瘢痕癌为研究对象,以10例正常皮肤组织为对照。免疫组织化学(SP法)技术检测Bcl-2蛋白的表达;以原位末端标记测定法(Tunel法)检测上述三种组织中的细胞凋亡情况。免疫组化结果应用图像分析软件计算阳性面积和平均光密度,Tunel结果计算凋亡指数。结果 (1)Bcl-2蛋白在正常皮肤表皮和病理性瘢痕上皮中分别仅见于基底细胞层少量表达、基底细胞层和极少量棘细胞层表达,均为阴性,在瘢痕癌组织中未见表达,为阴性,且其表达强度和表达水平在三组之间比较差异均无统计学意义(P<0.05);(2)Tunel结果示凋亡指数在三组间有差异,瘢痕癌组低于余两组,且与余两组间比较差异有统计学意义(P<0.05),但余两组间比较差异无统计学意义(P>0.05)。结论在瘢痕癌组织中Bcl-2蛋白呈阴性表达,其凋亡抑制作用有待进一步探讨。 Objective To explore the association of expression of Bcl-2 proteins with scar carcino- ma cell apoptosis.Methods (1)It was used that 15 cases pathological scar tissues and 20 cases skin scar carcinoma tissues to compare with 10 cases normal skin tissues.(2)The expression of Bcl-2 proteins were detected by the immunohistochemical (IHC) method of .SP.(3)The apoptosis po- sition these three organizations were detected by terminal deoxynucleotidyl transferase (TdT)-medi- ated dUTP nick end labeling(Tunel teehnology).(4)The images of IHC were calculated the positive area and the average optical density by image analysis software.The result of Tunel were calculat- ed the apoptotic index. The resulting data were used SPSS16.0 software for statistical analysis. Results (1)Bcl-2 showed negative expression in normal skin epidermis and pathological scar ep- ithelium,only a little expression in basal cell layer and prickle cell layer, negative expression in scar carcinoma tissue. The expression of intensity and levels differences were not statistically sig- nificant(P〉0.05) in the three groups.(2)Tunel test results showed that the apoptotic index difference in the three groups. Apoptotie index in the scar carcinoma group were lower than other two groups,and the difference were statistically significant (P〈0.05),however,the difference were not sta- tistically significant in other two groups.Conclusion Bcl-2 showed negative expression in scar carcinoma tissue. Its role of inhibition of apoptosis is to be further explored.
