摘要
为建立2型糖尿病大鼠模型,给予板蓝根水提物(WERI)及卡托普利干预,4周后分别检测各组大鼠血清中一氧化氮(NO)、一氧化氮合酶(NOS)、内皮素-1(ET-1)、单核细胞趋化蛋白-1(MCP-1)及细胞间黏附分子-1(ICAM-1)含量,并对主动脉进行苏木精-伊红(HE)染色后观察。结果表明:与模型组比较,卡托普利组及WEIR各剂量组大鼠血清NO含量及NOS活性显著上升,ET-1、MCP-1及ICAM-1含量显著下降。模型组大鼠主动脉HE染色结果显示主动脉内皮细胞和平滑肌细胞增生,且平滑肌细胞层数增多,排列紊乱、扭曲,而WEIR各剂量组及卡托普利组均得到改善。说明WEIR保护糖尿病大鼠的主动脉可能与增加血清中NO、NOS活性及降低血清中ET-1、MCP-1和ICAM-1含量有关。
To investigate the effect of water extract of Isatidis radix(WEIR) on type 2 diabetic rats.The model mentioned above was gavaged with WEIR or captopril to investigate the effect of WEIR on rats.After four weeks of administration,the concentration of nitric oxide(NO),nitric oxide synthase(NOS),monocyte chemotaxis protein-1(MCP-1),endothelin-1(ET-1)and intercellular cell adhesion molecule-1(ICAM-1) were measured.The pathological change of aortas was observated by hematoxylin eosin stain.The results showed that compared with the model group,the nitric oxide synthase activity,concentration of nitric oxide in captopril group and WEIR groups were increased,and the concentration of ET-1,MCP-1 and ICAM-1 were decreased.The aortic endothelial cells and smooth muscle cells of the model group were proliferated,and the smooth muscle cells of the model group were arranged disordered and distorted.The aortas of the WEIR groups and captopril group were significantly improved.It was concluded that WEIR could protect the aorta of diabetic rats,which may be related to the increase of serum NO,NOS and decrease of serum ET-1,MCP-1 and ICAM-1 level.
