伤科黄水治疗急性软组织损伤的药效学研究 被引量:19

摘要 目的:研究伤科黄水对急性软组织损伤的治疗作用。方法:将小鼠随机分为空白对照组,扶他林1.00 g/kg组,伤科黄水69.00 g/kg剂量组、34.50 g/kg剂量组、17.25 g/kg剂量组,每组15只。采用小鼠二甲苯耳肿胀模型和毛细血管通透性模型,考察伤科黄水的抗炎作用;采用小鼠热刺激致痛模型、小鼠醋酸扭体模型,以及小鼠甲醛致痛模型,考察伤科黄水的镇痛作用。采用重物高空击打法制备大鼠急性软组织损伤模型,将符合实验要求的大鼠随机分为空白对照组、模型对照组、扶他林0.50 g/kg组,以及伤科黄水34.50 g/kg剂量组、17.25 g/kg剂量组、8.63 g/kg剂量组,每组15只,连续外用给药5 d,观察组织损伤程度和病理学差异,测定血液流变学参数及炎性介质的变化。结果:伤科黄水69.00 g/kg剂量组、34.50 g/kg剂量组、17.25 g/kg剂量组可以明显减轻二甲苯对小鼠耳廓所致的肿胀,伤科黄水69.00 g/kg剂量组对醋酸引起的毛细血管通透性增加也有明显的抑制作用。伤科黄水各剂量对热刺激所致疼痛无显著镇痛作用,伤科黄水69.00 g/kg剂量组、34.50 g/kg剂量组对醋酸和甲醛致刺激性疼痛有明显镇痛作用,与空白对照组相比有显著性差异。本研究成功建立了大鼠急性软组织损伤模型,与模型对照组大鼠相比,伤科黄水34.50g/kg剂量组、17.25 g/kg剂量组大鼠在软组织损伤程度、病理形态变化及血液流变学方面有显著性的差异;伤科黄水8.63 g/kg剂量组、17.25 g/kg剂量组、34.50 g/kg剂量组大鼠局部损伤组织的前列腺素E2(PGE2)、白细胞介素-1β(IL-1β)的含量明显下降。结论:伤科黄水具有抗炎、镇痛的药理作用,对炎性渗出肿胀有显著的抑制作用,可用于治疗急性软组织损伤。 Objective: To investigate the effect of Shangkehuangshui on the acute soft tissue injury. Methods: The anti-inflammatory effect was observed in ear swollen of mice induced by dimethy benzene and capillary permeability test. The analgesic effect was observed by determining mice pain threshold in hot-plate test,writhing method and formaldehyde pain test. Acute soft tissue injury models in SD rats were established. Rats were divided into 6 groups and administered with diclofenac diethylamine emulgel and Shangkehuangshui for 5 days respectively.The injury degree of the soft tissue and pathologic changes were observed,and hemorheology indexes and inflammatory medium were measured. Results: The ear swollen of mice induced by dimethy benzene and capillary permeability were obviously relieved by Shangkehuangshui,and there were significant differencs compared with the control group( P 0. 05). Shangkehuangshui had no significant analgesic effect on the pain caused by heat,but it highly prolonged the pain threshold of mice in writhing method and formaldehyde pain test( P 0. 05). There were significant differences in the injury degree of soft tissue,pathologic changes and hemorheology of rats administered Shangkehuangshui compared with the model group( P 0. 05). Also,Shangkehuangshui decreased prostaglandin E2( PGE2) and interleukin-1β( IL-1β) in the damaged tissue,and there were significant differences from the model group( P 0. 05). Conclusion: Shangkehuangshui has the significant analgesic effect on the pain caused by the chemical stimulation,and obvious inhibitory effect on inflammatory extravasate in experimental animal models. Shangkehuangshui could be used to treat acut soft tissue injury.
