从AMPK介导的自噬研究淫羊藿总黄酮对自然衰老大鼠小肠屏障功能的调节作用 被引量:6

摘要 目的:从AMPK介导的自噬探讨淫羊藿总黄酮对自然衰老大鼠小肠屏障功能的调节作用。方法:SD大鼠按其不同发育阶段分为6月龄成年对照组,24月龄衰老组及淫羊藿总黄酮低、中、高剂量(10、20、60 mg/kg)组。用D-乳酸试剂盒检测各组血清D-乳酸含量;采用免疫组化检测各组小肠Claudin-1、Occludin、p-AMPK表达情况;Western blot检测小肠自噬相关蛋白Lc3Ⅱ、Beclin1表达情况。结果:与成年组比较,衰老组血清D-乳酸含量显著升高,小肠Claudin-1、Occludin、p-AMPK、Lc3Ⅱ、Beclin1表达显著降低(P<0.01);与衰老组比较,淫羊藿总黄酮中、高剂量组血清D-乳酸水平显著降低,小肠紧密连接蛋白与自噬相关蛋白的表达显著升高(P<0.01)。结论:淫羊藿总黄酮可能通过上调AMPK介导的自噬,增加肠上皮细胞间紧密连接蛋白的表达,从而改善衰老大鼠肠道黏膜屏障功能障碍。
