摘要
目的:观察灯盏乙素对双侧脑室注射Aβ25-35痴呆模型小鼠脑组织中差异蛋白表达的影响,筛选出药物调控的靶蛋白和信号通路,为下一步研究灯盏乙素抗痴呆的防治机制提供实验依据。方法:将昆明小鼠随机分为正常对照组、模型组和灯盏乙素组。用灌胃方法对灯盏乙素组进行给药(40 mg/kg)处理,历时90 d。用Morris水迷宫方法检测小鼠学习记忆能力;用同位素标记相对和绝对定量技术进行蛋白质组学分析。结果:灯盏乙素改善了模型小鼠下降的学习记忆能力,调控了6种靶蛋白和2条信号通路的表达。结论:实验获得了灯盏乙素改善Aβ所致的小鼠学习记忆能力损伤的重要蛋白质和通路信息。
Objective:To observe the effect of scutellarin on the expression of differential protein in brain tissue of dementia model mice which induced by bilateral intravantricular injected Aβ_(25-35),and to screen the target protein and signaling pathway of drug regulation,to provide experiment evidence in prevention and treatment mechanism of dementia.Methods:Kunming mice were randomly divided into normal control group,model group and scutellarin treatment group.The mice in scutellarin treatment group were treated with scutellarin by intragastric gavage(40 mg/kg)over a period of 90 d.The abilities of learning and memory were assessed by Morris Water Maze test.The analysis of proteomics were used by the technique of isobaric tags for relative and abosolute quantitation(i TRAQ).Results:scutellarin can improve the abilities of learning and memory in the mice of the model group.In addition,two pathways and six target proteins of drug regulation were found in this study.Conclusion:Information of important protein and pathway of the abilities damage of learning and memory induced by Aβ_(25-35) are obtained through the use of scutellarin.
