摘要
目的:探讨茶条槭果2种提取分离部位对小鼠H_(22)移植瘤的生长抑制作用及其机理。方法:通过小鼠左前肢腋下接种H_(22)小鼠肝癌细胞建立H_(22)荷瘤小鼠模型,腹腔给药10 d,观测肿瘤生长情况。计算抑瘤率,并通过试剂盒测量IL-6、IFN-γ、TNF-α、ALT、AST、BUN、CRE等指标来考察茶条槭果2种提取分离部位对小鼠H_(22)移植瘤的生长抑制作用。并进一步通过HE染色法观察肿瘤组织的病理改变,TUNEL法检测细胞凋亡情况。结果:茶条槭果2种提取分离部位均能降低荷瘤小鼠血清IL-6、TNF-α、INF-γ水平及CRE、BUN、AST、ALT水平,其给药组组织切片染色观察出坏死区域。结论:茶条槭果2种提取分离部位均能显著抑制肿瘤细胞生长、保护肝肾,提高免疫水平,通过诱导肿瘤细胞凋亡而发挥抑制肿瘤生长作用。
Objective: To study the inhibition and the mechanism of the two rough separation sites of Acer ginnala fruit in H22 tumor-bearing mice. Methods: H22 tumor-bearing mice were established by inoculating H22 mouse hepatoma cells in the left forelimb of the mice,and intraperitoneal administration drugs to observe 10 d,the tumor growth was also observed. The tumor inhibition rate was calculated,the levels of serum IL-6,IFN-γ,TNF-α,ALT,AST,BUN and CRE were measured by the kit to investigate the effects of the two rough separation sites on the H22 transplantation tumor growth inhibition. The pathological changes of the tumor tissues were observed by HE staining,and the apoptotic cells were detected by TUNEL method. Results: Two rough separation sites of Acer ginnala fruit could reduce the levels of IL-6,TNF-α and INF-γ,and decrease the levels of CRE,BUN,AST and ALT in the serum of H22 tumor bearing mice. Drugs groups were stained to observe the necrotic area. Conclusion: The results show that two rough separation sites of Acer ginnala fruit can significantly inhibit the growth of tumor cells,protect the liver and kidney,enhance the immune level,and inhibit the growth of tumor cells by inducing tumor cell apoptosis.
