摘要
目的:探索不同光质对桑黄菌丝生长及代谢产物累积变化规律。方法:分离、纯化、鉴定野生桑黄菌株;采用暗处理和白、蓝、绿、黄、红5种不同波长的光对桑黄菌丝进行照射培养,通过菌落直径、颜色变化及HPLC分析代谢产物累积变化和测定代谢产物对DPPH自由基清除活性变化,分析不同光质照射对桑黄菌丝生长及代谢产物累积影响。结果:分子生物学鉴定分离所得菌株为桑黄纤孔菌Inonotus sanghuang Sheng H.Wu,T Hatt.&Y.C.Dai。不同光质对该桑黄菌丝体生长有显著影响,其中暗培养、黄和红光处理生长速度及菌落形态上基本一致,但白、蓝和绿光处理生长抑制作用明显,菌丝看上去稀疏但菌落却厚实,并分泌大量黄色色素。HPLC分析结果表明,白、蓝和绿光处理20 d可显著增加代谢产物类型及丰度,其中苯乙烯基吡喃酮骨架类化合物Hypholomine B和Inoscavin A两个组分尤为显著,同时代谢产物对DPPH自由基清除活性也显著增加;蓝光处理后其胞内多糖却显著下降,而黄光处理则显著增加,其他处理组与暗培养组相当。不同光照处理时间各代谢产物HPLC图谱呈规律性变化,进一步说明光对桑黄代谢具有显著调控作用。结论:不同光质对桑黄生长、发育及代谢产物累积具有重要调控作用,暗培养有利于桑黄菌丝生长及提高生物量,蓝、绿光虽然对桑黄菌丝生长具有一定抑制作用,但可显著增加药效活性成分累积,因此,深入研究可为科学合理开发利用桑黄奠定基础。
Objective: To explore the effects of different light quality on the growth and metabolism accumulation in Inonotus sanghuang.Methods: Separation,purification and identification the wild fungus of Inonotus sanghuang,then the fungus was treated by dark,white,blue,green,yellow and red led light,the diameter,color change of the colony was recorded,and the change of the second metabolism and its free radical scavenging activity was analyzed by HPLC method and DPPH assay,respectively.Results: The fungus was identified as Inonotus sanghuang Sheng H.Wu,T Hatt. Y.C.Dai by molecular technique.Different light quality showed significant influence on the mycelial growth.The dark,yellow and red light treatment were the similar growth speed and colony morphology.But the white,blue and green light treatment manifested remarkable growth inhibition effect,with sparse hyphae,but the colony was thick and the solid with a large amount of yellow pigment secretion.Meanwhile,HPLC analysis indicated that the white,blue and green light treated for 20 d could dramatically increase the type and the content of the second metabolisms(Hypholomine B and Inoscavin A) and enhance the DPPH radical scavenging activity.However,the intracellular polysaccharide was significatly decreased by blue light treatment; in contrast,the yellow light treatment significatly increased intracellular polysaccharide,and other treatment was similar with the dark treatment group.HPLC fingerprint profiles of the second metabolisms changed regularly according to the different light quality with different time treatment,which reconfirmed that the light with remarkable regulation effected on the metabolism of Inonotus sanghuang.Conclusion: Different light quality had important regulation effected on the growth,development and second metabolism accumulation in Inonotus sanghuang.Dark culture condition is beneficial to the mycelial growth and biomass increasing,the blue and green light can increase the active components accumulation in spite of the growth inhibition effect.Hence,further research should be taken to pave the way for scientifically and rationally exploring Inonotus sanghuang.
