摘要
目的:建立HPLC同时测定酒山萸肉、乌梅、醋五味子、覆盆子、枸杞子、金樱子、木瓜和山楂中酒石酸、苹果酸、乳酸、柠檬酸、琥珀酸和富马酸含量的方法。方法:采用Agela Venusil MP-C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈∶0.01 mol/L磷酸氢二钠-磷酸缓冲液(p H=2.2)梯度洗脱,柱温25℃,流速0.8 m L/min,检测波长220 nm,进样量:10μL。结果:酒山萸肉、乌梅和醋五味子中均可检测到6种有机酸类成分,覆盆子、金樱子和木瓜中未检测到乳酸,覆盆子和枸杞子中未检测到酒石酸,山楂中未检测到富马酸。酒石酸、苹果酸、乳酸、柠檬酸、琥珀酸和富马酸在8种果实类药材中含量差异较大。结论:该方法简便、准确、快速,为药材中常见有机酸的含量测定提供方法依据。
Objective: To establish a HPLC method for the simultaneous determination of tartaric acid,malic acid,lactic acid,citric acid,succinic acid and fumaric acid,to determine the six organic acids in Corni Fructus,Mume Fructus,Schisandrae Chinensis Fructus,Rubi Fructus,Lycii Fructus,Rosae Laevigatae Fructus,Chaenomelis Fructus,Crataegi Fructus.Methods: Agela Venusil MP-C_(18) chromatographic column(250 mm × 4.6 mm,5 μm) was adopted for separation.The mobile phase was acetonitrile-0.01 mol/L disodium hydrogen phosphate buffer solution(p H 2.2) in gradient elution.The column temperature was 25 ℃,the flow rate was 0.8 m L/min,and detection wavelength was 220 nm,the injection volume was 10 μL to determine the contents of the organic acids in eight medicines.Results: Six organic acids could be determined in Corni Fructus,Mume Fructus,Schisandrae Chinensis Fructus,while lactic acid could not be determined in Rubi Fructus,Rosae Laevigatae Fructus and Chaenomelis Fructus; tartaric acid could not be determined in Rubi Fructus and Lycii Fructus,and fumaric acid could not be determined in Crataegi Fructus.The contents of tartaric acid,malic acid,lactic acid,citric acid,succinic acid and fumaric acid were in a great difference of eight herbs.Conclusion: This method is simple,accurate and rapid,which can provide the reference and quality control method for determination of common organic acids.
